中文摘要 | 欲對人類口腔癌細胞之生物特性有更深入之瞭解,必需先在實驗動物中,探尋實驗性口腔癌之各種生物特性。過去三十年間,許多學者已針對實驗動物之原發性口腔癌,進行詳盡之研究;對人類口腔癌之細胞特性,亦已有相當之報導。唯對原發性口腔癌演變成為極惡性的分化不全類上皮細胞癌之過程,即迄無報導。本研究曾自行建立實驗性之原發性口腔癌動物模式,並曾由此發展出可移植性的分化不全類上皮細胞癌之模式。故本研究室即運用此二模式之標本,進行口腔癌細胞在不同分化時期,其超顯微結構之病理研究,期能了解癌細胞在演變過程中之生物特性變化。由動物取得不同分化時期之口腔癌標本共30個,以穿透式電子顯微鏡技術,做系統性之偵測、記錄、及比較各類細胞之超顯微結構的病理變化與其生物特性之關聯。結果發現口腔癌細胞在分化程度愈差時,其病理變化愈嚴重且喪失規律性。若將分化不全之口腔癌細胞演化成分較佳之癌細胞,則其病理變化趨向緩和。此結果顯示,口腔癌細胞在各種不同之分化期間,其病理變化亦有不同,且此病理變化係與其生物特性呈直接關聯。 |
英文摘要 | Ultrastructural studies have been carried out on both DMBA-induced carcinomas of hamster buccal pouch and anaplastic carcinomas developed by serial transplantation of DMBA-induced buccal pouch carcinomas into abdomens of immunosuppressed neonatal hamsters. After two generations of serial abdominal transplantation, the anaplastic carcinomas were stable and could also be replanted to buccal pouches in non-immunosuppressed adult hamsters. The ultrastructural features of the transplanted anaplastic carcinomas were found to be generally similar to those of the primary well differentiated tumors, but the pathologic changes were more severe. Nuclei were irregular in size and shape and exhibited deep invaginations of nuclear envelope. Nuclear bodies were recognized. Nucleolar material was scattered and often condensed and marginated at the periphery of the nucleus. Markable decreases in cell attachments were observed with widened intercellular spaces and presence of villous-like configurations. Clumping of tonofilaments was also noted. These ultrastructural features are interpreted as indicator of increased protein synthesis and exchange of nuclear-cytoplasmic products. |
