英文摘要 | This study semen samples were collected from 67 boars 87 ejaculates. Boar semen collection was 5 days interval. All boars include 25 Landrace, 15 Duroc Jersey, 9 Large White, 8 Minnesota No. 2, 6 Pietrain, and 4 Hampshire boars. The semen was strained through a single layer of chessecloth to remove the gelatinous material and then mixed in a graduated glass-stopped cylinder by inverting three or four times. Samples of approximately 30 ml. each were taken with a 10 ml. of pipette immediately after mixing the ejaculate, from upper, middle, and buttom of the cylinder, and placed in 50 ml. glass-stopped cylinder, corked, and stored at 15℃. Semen concentration were counted by Thomas' hemocytometer method (each samples count three times) and photo-colorimeter method (each samples test two times). The poto-colormeter method is tested using No. 623 Blue green Ilford filter of EEL-morder 2 portable Colormeter with maximum transmission at 490 mμ. Each samples were diluted at rate of 1:9, it means by pipetting 9 ml. of a 2.9% sodium citrate dihydrate solution into a clean absorption cell and adding 1 ml. of semen with another pipette. After adding of semen to the 2.9% sodium citrate dihydrate solution, part of the solution was drawn back into the pipette three times to rinse the pipette. The standard referance tube contained 10 ml. of 2.9% sodium citrate dihydrate. The average of all samples sperm concentration is 373.1×10?/ml. (45.5-859.0×10?/ml.) for Thomas' hemocytometer method, and the average of reading pointer is 3.42 (0.32-6.68) for colormeter method. The result of two methods have highly linear correlation (r=0.966). The regression equations for estimating sperm concentration (Y) (in millions per ml.) in boar semen, Y=121.82 X-44.07, where x equals Colorimeter reading. (with 1:9 dilution rate). |
