| 中文摘要 | 血液尿酸的升高不只和痛風的發生有密切相關,近年來的研究亦指出高尿酸血症和腎臟疾病、心臟疾病以及一些慢性發炎的發生率有著顯著的正相關,因此尿酸檢驗在一般健康檢查當中爲不可或缺的一個項目。爲了使得每個實驗室都可以以簡便、快速、便宜的方法測尿酸,本實驗室參考文獻自行配置可用於96孔微量滴盤上之尿酸檢驗試劑,並使用通過國際認證之尿酸標準品,不論尿液或血清檢體,手工尿素酶-過氧化酶酵素法的結果和Hitachi機台得到之結果相關性佳(R=0.98),可偵測的範圍0.3~70.6 mg/dL,檢驗的靈敏度可達0.3 mg/dL;檢驗不精密度當日爲1.6~1.8%,每日之間爲1.7%~2.5%。而價錢上的比較,手工方法每個檢驗所需成本僅爲0.6元。我們相信使用尿酸微量滴盤之檢驗方法對於未來預測第2型糖尿病患者族群發生心血管疾病的風險將有所助益。 |
| 英文摘要 | Uric acid, a degradation product of purine, is a well-known marker for gout. Recent reports have also indicated that uric acid is an independent risk marker for cardiovascular disease. Conceivably, a simple and rapid procedure for measuring uric acid concentration is in great demand. We have modified an enzymatic method for uric acid that can be used for quantifying uric acid in either serum or urine utilizing 4-aminoantipyrine/TOOS system and using both enzymes of uricase and horseradish peroxidase. An excellent correlation with correlation coefficient equals to 0.99 was found between our microplate procedure and that of the Hitachi auto-machine. The concentration range of our microplate method was determined from 0.3~70.6 mg/dL with a sensitivity of 0.3 mg/dL. The within-run imprecision and between-run imprecision are found to be 1.6%~1.8% and 1.7%~2.5%, respectively. Our method is relatively inexpensive. It is estimated to cost only NT$0.6 /test. We believe that our simple microplate procedure is useful for assessing the risk of developing cardiovascular disease in patients with type 2 diabetes. |
