| 中文摘要 | 本研究自土壤中分離出黑殭菌 ( Metarhizium anisopliae ) CCC 901 菌株,將其孢子懸浮液濃度配製成 107 conidia/ml,於室內 25℃及高相對濕度環境下,對甘藍菜上黃條葉蚤 ( Phyllotreta striolata )、小菜蛾 ( Plutella xylostella )及偽菜蚜 ( Lipaphis erysimi ) 等蔬菜重要害蟲進行致病力試驗,結果顯示接種孢子懸浮液 7 天後,感染 致死率皆可達 79%以上,顯示此菌株具有寄主範圍廣泛之特性。將 CCC 901 菌株以六種不同穀類基質進行固態培養 (solid cultivation) 試驗,結果顯示該菌株在白米基質上培養至第 14 天的產孢速率最快且產孢量最多,可達 2.7×108 conidia/g。於暗環境下利用四種液態培養基震盪搖瓶進行黑殭菌 CCC 901 菌株之液態培養 (liquid cultivation) 試 驗,結果顯示培養 7 天後,其於 Sabouraud Maltose Agar Yeast (SMAY) 培養基中產生的芽生孢子量最高,可達 4.2×107 blastospore/ml 。 再 將 上 述 經 四 種 液 態 培 養 基 產 生 的 黑 殭 菌 CCC 901 芽 生 孢 子 懸 浮 液 稀 釋 成 107 blastospore/ml,分別接種於白米基質上進行雙相培養 (diphasic cultivation) 試驗,結果顯示培養 14 天後,以 SMAY 之芽生孢子懸浮液接種的產孢量最高,可達 7.3×108 conidia/g。黑殭菌 CCC 901 菌株對多種蔬菜害蟲具優異的感染 效果,且室內初步量產之雙相培養結果,可供做往後菌株量產及田間應用之參考。 |
| 英文摘要 | In this experiment, Metarhizium anisopliae CCC 901 strain was collected and screened from from soil. In addition, the pathogenicity of the the conidial suspension of CCC 901 strain (107 conidia/ml) as the inoculum under high relative humidity in the laboratory (25℃) against other pests such as Phyllotreta striolata ,Plutella xylostella , and Lipaphis erysimi on cabbage was conducted. The results showed that the average mortality was above 79% at 7 days after inoculation, indicating that CCC 901 possess broad-host pathogenicity. Six different kinds of grains were used for mass spore production of CCC 901 strain by solid cultivation. The results showed that the strain had the fastest sporulation rate and the largest spore production at 14 days after cultivation for rice substrate, which could reach 2.7×108 conidia/g. In addition, CCC 901 liquid cultivation was performed in a dark environment using four kinds of liquid medium with rotatory shaking. The results showed that the highest blastospore concentration of CCC 901 strain reached 4.2×107 blastospore/ml for Sabouraud Maltose Agar Yeast (SMAY) liquid medium at 7 days after cultivation. Besides, the CCC 901 blastospore suspensions produced by the above four liquid medium after 7 day’s cultivation were diluted to 10 7 blastospore/ml and inoculated on the rice substrate for diphasic cultivation test. The results showed that the highest spore concentration of CCC 901 strain reached 7.3×10 8 conidia/g inoculated with SMAY blastospore suspension at 14 days after cultivation. This study suggests that M. anisopliae CCC 901 strain possesses excellent infective effects on a variety of vegetable pests, and the diphasic cultivation results of CCC 901 in the laboratory can be used for mass spore production and field application of them in the future. |
