中文摘要 | 本研究目的旨在探討運動介入對老齡大鼠阿基里斯肌腱斷裂後修復的影響。實驗方法以48隻2個月大(8週)及18月大(72週)之Spraque-Dawley品系公鼠為對象。實驗為期84天。隨機分『幼齡控制組』(自由活動)、『幼齡運動組』、『老齡控制組』(自由活動)、『老齡運動組』,共4組;先將大鼠右後腳阿基里斯肌腱切斷使其受傷。控制組飼養在籠內任其自由活動;運動組介入方式為游泳+走路,術後第5天開始進行游泳運動,第14天改為走路運動,運動介入外的時間均任由大鼠在籠內自由活動。分析受傷後第84天,每組隨機取3隻大鼠以組織切片、染色觀察膠原蛋白生長情形;每組隨機取9隻大鼠進行生物機械性能測試。結果:一、受傷後28天各組運動組的AFI (Achilles tendon Functional Index)值顯著比各組控制組高,傷後84天週4組間則無顯著差異。二、膠原纖維結構中4組均出現波紋結構,但控制組的排列較紊亂,膠原蛋白種類比例(collagen type proportion Rate, CTR)方面4組均無顯著差異。三、傷後84天4組的受傷肢肌腱直徑(Achilles Tendon Diameter, ATD)皆顯著大於正常肢。四、運動組受傷肢肌腱斷裂強度(Breaking Stress, BS)顯著大於正常肢。五、受傷肢與正常肢之間的肌腱拉伸應力(Tensile Stress, TS)達顯著差異。結論:不論幼齡或老齡,運動皆會使傷後第28天步態恢復較快,而運動組相較於控制組在傷後第84天雖然有較高的斷裂強度,各組受傷肢拉伸應力卻仍低於正常肢。 |
英文摘要 | The aim of this study is to examine the effects of exercise intervention on ruptured Achill’s tendon recovery of aged rats. Methods: There are 48 young (8-week-old) and aged (72-week-old) Spraque-Dawley (SD) rats during an 84-day experimental period. All rats are randomized into four groups: young control, young exercise, aged control, and aged exercise. All data are expressed as mean ± standard deviation. The parameters are compared using a three-way mixed ANOVA measure. The significant level is set as α=.05. It is expected that the intervention of exercise can benefit the recovery of the ruptured tendon. Additionally, the differences in ruptured tendon recovery between young and aged rats are examined in this study. Result: AFI values: Both two exercise groups were significantly better than control group of AFI at 28^(th) post injury. And there were no significantly at 84^(th) post injury. Collagen growth: All group was form crimp to collagen structure. No found significantly between the four groups of collagen type proportion at 84^(th) post injury. Tendon diameter: No found significantly to normal limbs and injury limbs to tendon diameter between the four groups at 84^(th) post injury, but normal limbs were significantly smaller than injury limb to tendon diameter in each groups. Tendon breaking strength: There were significantly to normal limbs and injury limbs to tendon breaking strength between the exercise groups, but normal limbs were significantly smaller than injury limbs to tendon breaking strength of exercise group at 84th post injury. Tendon tensile stress: No found significantly to normal limbs and injury limbs to tendon tensile stress between the four groups, but injury limbs were significantly smaller than normal limb to tendon tensile stress in each groups. Conclusion: Regardless of young or aging, all the movements recovered faster on the 28th day post injury. |
