中文摘要 | 表現F4 纖毛的腸毒素大腸桿菌(ETEC)為常見的腸道病原菌,會引起新 生仔豬與離乳豬下痢及離乳後豬隻的水腫病。黏蛋白基因 4(MUC4)、黏蛋白基 因 13(MUC13)和轉鐵蛋白受體基因(TFRC)被認為是腸毒素大腸桿菌 F4ab/ac 抗性的候選基因,然而,目前尚缺乏有關選拔豬隻腸毒素大腸桿菌 F4ab/ac 抵抗力 對其生長性能可能影響之報告,本研究之目的在探討藍瑞斯、約克夏和杜洛克等 臺灣主要西方品種豬隻腸毒素大腸桿菌 F4ab/ac 抵抗力相關基因單核苷酸多態型 (SNPs)對生長性能之影響。研究共使用 1480 頭於 2005 年 1 月至 2007 年 12 月 期間以隔離早期離乳進豬之中央檢定公豬(1057 頭杜洛克、317 頭藍瑞斯和 106 頭約克夏),以聚合酶連鎖反應限制片段長度多態型分析法決定MUC4、MUC13 和 TFRC 基因 SNPs 之基因型。研究結果顯示 TFRC 在所調查的臺灣種豬群並未發 現多態型;對腸毒素大腸桿菌 F4ab/ac 具易感性之 MUC4-C8227G 之 G 交替基因 頻率在杜洛克、藍瑞斯和約克夏分別為 0.02、0.46 和 0.51;MUC13-A908G 基因具 易感性之 G 交替基因頻率在此三品種豬隻分別為 0.50、0.71 和 0.62。統計分析之 結果顯示 MUC4 及 MUC13 基因之SNPs 對杜洛克和約克夏之生長性狀無顯著效應 (P > 0.05);MUC4 基因 SNP 會顯著影響藍瑞斯豬之到達 110 kg 日齡、150 日齡 體重及其背脂厚度。本研究結果顯示以 MUC4 及 MUC13 基因 SNPs 選拔腸毒素大 腸桿菌 F4ab/ac 抗病力將不會影響杜洛克和約克夏豬隻之生長性能,但可能影響藍 瑞斯豬之生長性能。 |
英文摘要 | Enterotoxigenic E. coli (ETEC) expressing F4 fimbriae is a common porcine enteric pathogen, causing diarrhea in new born and weaning pigs, and edema disease in pigs after weaning. The mucin 4 (MUC4) , mucin 13 (MUC13) and transferrin receptor (TFRC) genes have been proposed as candidate genes for the disease resistance to ETEC F4ab/ac. However, there were still few reports on the possible effect of selecting resistance to ETEC F4ab/ac on the growth performance in pigs. The purpose of this study was to investigate the effect of single nucleotide polymorphisms (SNPs) in ETEC F4ab/ac resistance-related genes on the growth performance in exotic purebred pigs in Taiwan. For evaluating the effect of selecting SNPs on growth performance traits, 1,480 centrally performance tested purebred boars (1057 Duroc, 317 Landrace, and 106 Yorkshire) from January 2005 to December 2007 were used. The genotypes of MUC4, MUC13 and TFRC SNPs were determined by polymerase chain reaction based restriction fragment length polymorphism. The results indicated that there was no polymorphism detected in TFRC gene in the pig population surveyed. The allelic frequencies of the ETEC F4ab/ac susceptive G allele in MUC4-C8227G in Duroc, Landrace, and Yorkshire pigs were 0.02, 0.46, and 0.51, respectively. The allelic frequencies of G allele in MUC13-A908G in the three breeds of boars were 0.50, 0.71, and 0.62, respectively. Results of statistical analysis revealed that there was no significant effect of the SNPs in MUC4 or MUC13 on the growth performance of Duroc and Yorkshire pigs (P > 0.05). The SNP in MUC4 gene showed significant effect on days of age adjusted to 110 kg body weight and body weight at 150 days of age in Landrace (P< 0.05). The SNP in MUC13 significantly affected BF in Landrace (P < 0.05). The results of this study suggested that selecting disease resistance to ETEC F4ab/ac by SNPs in MUC4 and MUC13 did not affect the growth performance in Duroc and Yorkshire pigs, but may affect the growth of Landrace pigs. |
