中文摘要 | 本試驗主要探討細胞分裂素對石蓯蓉葉片誘導不定芽的影響,以建立石蓯蓉組織培養量產技術。以石蓯蓉葉片為培植體,培養於含有0.5 mg L^(-1) IBA + 1 mg L^(-1) BA的固體MS培養基8週後,不定芽誘導率為100%,每培植體可誘導之不定芽數最多為41.5個。將誘導的不定芽培養於含3000倍的Hyponex^® No. 5及0.1%活性炭之固態MS培養基中。結果顯示不定芽長度、根長、葉數及根數,分別為45.8 mm、60.5 mm、16.5及6.2個,可促進不定芽生長及發根。2個月後,將小苗移植到戶外,種植在培養基(泥碳土:蛭石:土= 2:1:1)中,小苗的存活率高達91.7%。本試驗建立石蓯蓉組織培養量產技術,除可提供種苗繁殖技術外,未來亦可應用於其族群復育。 |
英文摘要 | The potentialities of micropropagation from leaf explants of Limonium sinense (Girard) Kuntze in vitro were investigated. Leaf explants were cultured on Murashige and skoog's basal medium (MS) supplemented with different concentrations of 6-Benzyladenine (BA), 2-ip, Kinetin and Thidiazuron (TDZ) individually and in combination with a fixed concentration of 0.5 mg L^(-1) indole-3-butyric acid (IBA) for adventitious shoot induction. Direct adventitious shoot regeneration occurred on the most of the media. High shoot formation (100%) was obtained with 1 mg L^(-1) BA in combination with 0.5 mg L^(-1) IBA which produced 41.5 shoots per explant. The adventitious buds were induced in culture medium containing 3000X Hyponex^® No.5 and 0.1% activated charcoal in solid MS medium. The results showed that the adventitious bud length, root length, leaf number and root number were 45.8 mm, 60.5 mm, 16.5 and 6.2, respectively, which could promote adventitious bud growth and rooting. After 2 months, plantlets were transplanted to the outdoors and planted in the culture medium (peat moss: vermiculite: soil = 2: 1: 1), the survival percentage of plantlet up to 91.7%.. The protocol for micropropagation of Limonium sinense can be used as propagation techniques, moreover, it is useful for conversation purposes of endangered statice species. |
