中文摘要 | 阿里山十大功勞為國內重要珍貴藥用植物。本實驗材料取自嘉義縣阿里山森林遊樂區的阿里山十大功勞根菌結合體及種子;在根部形態觀察部份,利用光學顯微鏡進行染根觀察,可於根菌結合體內發現暗色隔膜菌絲及透明微菌核等構造;在菌株誘導、分離與純化部分,共計純化出2株內生菌(MoAL2和MoAL5),經分子生物定序比對及傳統形態特徵鑑定,此2株菌株分別隸屬於Hymenoscyphus menthae和Cladophialophora chaetospira。利用這2株菌株與阿里山十大功勞無菌苗進行半合成試驗,接種3個月後顯示,接種苗生長正常,無產生病徵現象。因此,本研究證實,阿里山森林遊樂區之阿里山十大功勞根菌結合體係屬暗色隔膜內生菌;其所分離純化菌株MoAL2和MoAL5應屬台灣新紀錄種,且能與阿里山十大功勞種子苗共生。 |
英文摘要 | Mahonia oiwakensis (Alishan mahonia) is an important medicinal plant in Taiwan. In this study, the root associations and seeds were collected from Alishan recreational park. Under light microscope, the stained root showed the melanized septate hyphae and hyaline microsclerotia in root cells. Two endophytes ( MoAL2 and MoAL5 ) were isolated from the root spectrums, and through traditional and molecular analysis, the two strains were found to belong to Hymenoscyphus menthae and Cladophialophora chaetospira. Three months after being inoculated with the two strains, the MoAL2 and MoAL5- inoculations showed no pathogenic symptoms in the semi-aseptic synthesis experiment. This study confi rmed that the root-fungus association of Mahonia oiwakensis belonged to dark septate endophyte. The two endphytes (MoAL2 and MoAL5) were newly recorded in the fungal flora of Taiwan, and both could synthesis with Mahonia oiwakensis. |
