中文摘要 | 牛樟扦插繁殖為國內重要牛樟造林木來源。目前本研究室已證實牛樟根菌結合體具有暗色隔膜內生菌之構造,但菌株與牛樟扦插苗之合成關係尚未釐清。材料取自於嘉義縣阿里山鄉達邦地區的牛樟根菌結合體及嘉義林區管理處所提供的扦插繁殖苗;在菌株分離部分,本試驗分離純化出2株內生菌,經分子生物技術比對,及傳統形態特徵鑑定,此菌株應屬於世界新種。利用這2株菌株與牛樟扦插苗進行半合成試驗,於接種6個月後顯示,接種苗生長旺盛,其根系利用光學顯微鏡進行染根觀察,皆可於根菌結合體內發現微菌核之構造。因此,本研究證實,菌株CkDB2和CkDB5為暗色隔膜內生菌,亦能與牛樟扦插苗於共生後,形成暗色隔膜內生菌之菌根型態。 |
英文摘要 | Cutting is an important propagation for plantation of "Cinnamomum kanehirai" in Taiwan. Previous study demonstrated that "C. kanehirai" could form dark septate endophyte in the field, but the study didn't indicate which strain could form microsclerotia with "C. kanehirai" cuttings. In this study, the root spectrums were collected from Dabang in Chiayi county, and "C. kanehirai" cuttings were supplied by Chiayi Forest District Office. Two endophytes were isolated from the root spectrums, and through traditional and molecular analyses the strain CkDB2 and CkDB5 were found to belong to a new addition in the fungal flora of Taiwan. In semi-aseptic synthesis experiment, these 2 endophytes (CkDB2 and CkDB5) could form microsclerotia in root associations by light microscopy. The result showed that these two endphytes were dark septate endophytes, and they could form a typical mycorrhiza (dark septate endophyte) with "C. kanehirai" cuttings. |
