中文摘要 | α-galactosidase (α-半乳糖??)是一種可催化水解α-半乳糖?寡糖之α-1,6鍵結的酵素,在食品及醫學等方面皆有其重要性。本研究利用含有 4-Methylumbelliferyl-α-D-Galactopyranoside (4MU-α-Gal)的洋菜平板針對22株乳酸株進行篩選,其中Lactobacillus fermentum L9, L.reuteri L22, L. crispatus L3, L. casei L19 及L. delbrueckii subsp. delbrueckii L7 等5株菌具有α-Gal酵素活性。進一第以p-NGP為基質利用光學定量分析法檢測α-Gal酵素活性,結果與4MU-α-Gal洋菜平板之篩選結果一致。應用L. reuteri L22菌株於α-半乳糖?類寡糖之水解反應,對於棉子糖及水蘇四糖之分解效率分別可達73%及64%。 |
英文摘要 | Alpha-galactosidase catalyses the hydrolysis of the α-1,6 linked galactoside fromα-galactose oligosaccharides. This enzyme has been used in several important fields such as food science and medicine. In this study we utilized 4-methylumbelliferyl-α-D-Galactopyranoside (4MU-α-Gal) agar plate to screen theα-galactosidase-positive bacterial strains from 22 identified lactic acid bacteria and five strains, Lactobacillus fermentum L9, L. reuteri L22, L. crispatus L3, L. casei L19 and L. delbrueckii subsp. delbruecki L7, showed positive results. Theα-galactosidase activities of those five strains were futher confimed by spectrophotometric analysis using p-NPG as the substrate. Hydrolysis ofα-galactose oligosaccharides by L. reuteri L22 was determined by HPLC analysis and the results demonstrated that 73% of raffinose and 64% of stachyose in solution were eliminated respectively. |
