中文摘要 | 本研究之目的在探討菜鴨(Tsaiya duck: Anas platyrhynchos var. domestica)卵巢濾泡細胞層內類固醇生成及其相關蛋白與酵素表現量之關聯性。收集菜鴨排卵前濾泡F1、F2、F3及F4/F5之粒性及膜性細胞層,分析類固醇含量與類固醇生成急性調控蛋白(steroidogenic acute regulatory protein, StAR)、P450膽固醇側鏈截切酵素(P450 cholesterol side-chain cleavage enzyme, P450scc)、3β-羥化類固醇去氫酶(3β-hydroxysteroid dehydrogenase, 3β-HSD)與芳香化酶(P450 aromatase, P450 arom)的表現變化。由類固醇含量結果顯示,以孕酮(P₄)含量而言,粒性細胞層高於膜性細胞層,且粒性細胞層內以F1含量最高;此外,排卵前濾泡的粒性細胞層的P4含量濾泡的逐漸成熟而增加(P<0.05)。然而,濾泡細胞層內睪固酮及雌二醇的含量,則以膜性細胞層高於粒性細胞層性細胞層內的睪固酮含量在F1-F4/F5濾泡並無顯著差異。菜鴨排卵前濾泡粒性細胞層的StAR、P450scc與3β-HSD表現量都是以F1最高,且此三種蛋白質的表現量隨濾泡的逐漸成熟而增加;P450arom在不同時期濾泡中的表現量則無差異。在F1至F5濾泡膜性細胞層中,發現StAR、P450scc與 3β-HSD表現量並沒有差異;P450arom則是在F4/F5的表現最高於其他時期的濾泡。綜合以上結果顯示,(1)菜鴨排卵前濾泡粒性細胞層內生成之P₄與其盯關之蛋白與酵素(StAR、P450scc與3β-HSD)的表現量呈正相關;(2)F1濾泡藉由增加與P₄生成相關之蛋白(StAr)與酵素(P450scc和3β-HSD)的表現量,大量生成P₄,進而誘發排卵的發生;(3)F2-F4/F5膜性細胞層含有高量的T,但其相關蛋白與酵素的表現量並未增加。 |
英文摘要 | The purpose of this study was to investigate the correlation of steroidogenesis with the expression of steroidogenic enzymes in preovulatory follicles of Tsaiya duck (Ana platyrhynchos var. domestica), Preovulatory follicles (F1-F5) were collected and the granulosa and theca layers o follicles were isolated. The concentration of progesterone (P₄), testosterone (T) and estradiol (E₂) were detected with EIA method. The expression of StAR (steroidogenic acute regulatory) protein. P450scc (P450 cholesterol side-chain cleavage enzyme), 3β-HSD (3β-hydroxysteroid dehydrogenase) and P450arorn (P450 aromatase) were measured. The results showed that progesterone content in the granulosa layer was higher than in the theca layers, especially in F1 and it was increased gradually with the maturity of the follicle. In contrast, the T and E₂ contents of theca layers were higher than granulosa layers. The T and E₂ contents of the F4/5 layer were the highest in different stages (P<0.05), but there was no difference in granulosa layers obtained from different stages of preovulatory follicles. The expression of StAR, P450scc and 3β-HSD was the highest in F1 granulosa layer and the expression levels gradually increased with the maturity of follicles. However, the expression level of P450arom showed no significant difference in different stages of granulosa layer development. In theca layers of preovulatory follicles, expression of StAR, P450scc and 3β-HSD showed no difference, hut the P450arom expression of F4/F5 was significantly higher than in other stages. In brief, it could be concluded that: (1) the biosynthesis of P₄ is positively correlated with the expression of key steroidogenic enzymes (StAR, P450scc and 3β-HSD) in preovulatory follicles of Tsaiya duck; (2) the F1 granulosa layer mainly secretes P₄ by increased enzymatic activity and expression of StAR, P450scc and 3β-HSD that induce ovulation; (3) the F3 and F4/F5 of theca layers produce a high level of T, but the expression level of StAR was not different. |
