中文摘要 | 研究目的:以代謝體學途徑結合多變數統計,對藥材進行全指紋圖譜分析,再加上藥理活性試驗資料的整合,能補充傳統品質管制方法中,僅以中藥材主要成分的分析角度所忽略之多成分組成與複雜生物活性間的關係的不足。並以主成分分析法(PCA)進行統計分析所繪製之落點圖,可作為中藥炮製品質管制監測分析的補充參考。 研究方法:利用 NMR進行不同炮製黃精樣品之指紋圖譜,配合主成分分析法統計,尋找其關連性與差異性。不同炮製的黃精樣品經過萃取、冷凍乾燥後進行樹突細胞(Dendritic cell)與周邊血液單核細胞(PBMCs)實驗之免疫活性測定。 結果與討論:一、在 NMR結合 PCA的統計分析落點圖中,臺灣市場中大部份中藥房所出售之製黃精與未炮製之黃精在 PCA的標準落點圖差距相當遠,其代表的意義為這些製黃精市場品都經過蒸製處理,但少數樣品卻與其他樣品不同,值得注意。二、蒸製黃精與未炮製的原材料之 PCA比較,發現黃精ㄧ經過蒸製處理後,立即與未炮製生黃精有明顯的差異,但是發現蒸製五次至九次的結果是相似的,表示五蒸五曬後的黃精化學成分品質已趨於一致與穩定。三、多次炮製的黃精樣品對 in vitro之樹突細胞無明顯的免疫活性與毒性。四、大鼠餵食之 in vivo實驗中,餵食製黃精的大鼠血液之 CD4濃度上升,而未炮製組則下降。同樣地,餵與黃精之大鼠的 CD8濃度皆有下降的趨勢。而餵與製黃精之大鼠的 CD4/CD8比值遠大於餵食未炮製黃精大鼠的比值,更大於空白對照組。而在免疫抑制之大鼠實驗方面,不論餵食黃精萃取物與否,對 CD4的濃度都有增加的趨勢,以生黃精效果最佳,CD4/CD8比值也以生黃精效果最好。顯示黃精的免疫作用必須透過動物體內代謝後才能產生刺激提升作用。 |
英文摘要 | AIM: Metabolomics approach in combination with multivariate statistics has been applied on the fingerprint of Chinese herbal materials, and linking with the patterns of pharmacological and bioactive tests is able to provide the profile and correlations between complex compounds and bioactivity due to synergic or opposing effect in traditional Chinese medicine. It can replenish the insufficiency of classic quality control methods based on the analysis of only major components. NMR combined with principal component analysis (PCA) to fingerprint the profile and standard plot of Chinese herbal materials is able to be the reference of ultimate QA/QC of Chinese materials processing. METHOD: In order to look the relation and difference, the fingerprints of different processed Rhizoma Polygonati were measured using NMR in combination with PCA. These samples in different processed were applied to investigate on the immune activities in dentritic cells and Peripheral blood mononuclear cells (PBMCs). RESULTS & DISCUSSION: 1. In the score plot of PCA combined with NMR, the difference between processed and non-processed Rhizoma Polygonati materials from Taiwan markets are quite big. It means these materials of Rhizoma Polygonati have been processed well done, even some cases were still out of score. 2. From the results of PCA, the first processed material was quite different from the non-processed ones. The PCA result indicates it was not much significant difference between 5 cycles and 9 cycles. It means the property will be steady after 5 cycles of steamed processing in Rhizoma Polygonati materals. 3. There was no immune stumilation and cyctotoxic effect on dentritic cells from processed Rhizoma Polygonati. 4. In the in vivo rats experiment, it indicated the concentration of CD4 was increased on the feeding of processed Rhizoma Polygonati extract and reducesd on the non-processed one, respectively. The ratio of CD4/CD8 casued by processed Rhizoma Polygonati was much higher than the control one. It means the processed Rhizoma Polygonati has the effect of immunesystem stumilation for normal rat. However, the concentration of CD4 had been increased significantly by the feeding of Rhizoma Polygonati extract on the immune-inhibited rats which induced by detramethasone. In addition, the ration of CD4/CD8 had been inhanced by non-processed Rhizoma Polygonati extract for immune-inhibited rats. |
