中文摘要 | 研究目的:本研究計畫主要是評估中藥材輻射滅菌前的最適合的乾燥處理方式,藉以降低以鈷六十加馬射線對中藥材進行照射過程中所產生輻射裂解的現象,避免影響中藥材化學成分。除了能提供國內外中醫藥業一種簡單實惠的滅菌及包裝貯存方式,更能對中藥材達到控管品質的要求。 研究方法:我們將以不適合室溫貯存的二十五種中藥材為研究對象,中藥材先經 50℃乾燥處理後,再以 3、6、9 kGy三種不同劑量的加馬射線照射後,經萃取後以 HPLC進行化學成分分析,以 GC/MS進行有機氯農藥檢測,以及利用平板檢測法進行菌量檢測。篩選出對此二十五種中藥材最適合的乾燥處理方式,最適合的加馬射線輻射滅菌劑量,再整合先前的研究成果,期能建立不同中藥材輻射滅菌最佳參數的資料庫,有利於強化中藥用藥安全。 結果與討論:此二十五種中藥材經萃取後以 HPLC進行化學成分分析,結果顯示: 9 kGy的加馬射線照射不會影響防風、蓮子、黃耆、台烏、延胡索片、黑棗、麻黃、玄蔘、香附子、何首烏、芡實、生地榆、佛手柑、蒿本、秦艽、田七、桃仁、遠志、青皮、獨活、天麻等二十一種藥材的化學成分。紅棗、木香及板藍根等藥材的安全劑量 6 kGy,何首烏的安全劑量 3 kGy。再利用平板檢測法進行菌量檢測,結果顯示貯存三個月的藥材菌量檢測結果顯示台烏、何首烏、前胡、芡實、木香、地榆、佛手柑、蒿本、秦艽、田七、桃仁、板藍根、獨活、天麻與等十四種中藥生材以 3 kGy的加馬射線輻射照射即可達滅菌的效果。黃耆、紅棗、延胡索片、麻黃、玄蔘、香附子、遠志、青皮與蓮子等九種中藥生材須以 6 kGy照射劑量才可達滅菌的效果,防風則需 9 kGy照射劑量才可達滅菌的效果,但是 9 kGy加馬射線照射劑量並無法使黑棗完全滅菌。貯存九個月的藥材菌量檢測結果顯示前胡、芡實、木香、地榆、佛手柑、蒿本、秦艽、田七、桃仁、天麻等十種中藥生材以 3 kGy的加馬射線輻射照射即可達滅菌的效果。台烏、何首烏、板藍根、獨活、黃耆、紅棗、玄蔘、香附子、遠志與蓮子等十種中藥生材須以 6 kGy照射劑量才可達滅菌的效果,防風、青皮、麻黃與延胡索片則需 9 kGy照射劑量才可達滅菌的效果,但是 9 kGy加馬射線照射劑量並無法使黑棗完全滅菌。以 GC/MS對其中產菌量較少的八種藥材進行有機氯等 208種農藥檢測,結果顯示並無農藥的殘留。 |
英文摘要 | AIM: The aim of this study is to reduce the radiolysis effect by utilizing drying process before 60Co gamma radiosterilization, and evaluate the appropriate process for the drying treatment. The application of 60Co gamma rays to decontaminate Chinese medicines could associate with certain problems. METHOD: In the previous study showed that the drying process at 50℃for 48 hours before irradiation not only could inhibit the bacterial growth, it also could decrease the radiolytic effect. Therefore, the drying process can decrease the irradiation dose for Chinese medicines radiosterlization. Twenty five Chinese medicines, which cannot be stored at room temperature, are dried at 50℃ for 48 hours before irradiation. Samples are treated using irradiation at doses of 3, 6 and 9 kGy, respectively. Characterizations of chemical compounds are performed by HPLC. The efficiency of radiosterization is determined by the growth of bacterial colonies from an LB mixture of each herbal medicine. Organochlorine pesticides are analyzed by GC/MS. The data of optimal drying temperature and irradiation dose for various Chinese medicines can improve the economic value and safety of Chinese medicines. RESULTS & DISCUSSION: After extraction, the chemical composition of 25 Chinese herbal medicines was analyzed by HPLC. The flat-flask detection method was used for bacteria detection. The results showed that nearly all the medicines were sterilized after irradiation treatment of 3kGy-γ ray. Compounds and functional groups in these herbal medicines were not changed under the dosage. Three kGy of gamma-ray can irradiosterlize the Radix Peucedan, Polygonum and other 12 kinds of Chinese medicines. For Astragalus, Lotus and other 7 kinds of Chinese medicines, 6 kGy was effective for irradiosterlization. The pesticide residues were analyzed by GC-MS. The results showed no detective pesticide on the 8 Chinese medicines which had less bacteria colony formation. |
