中文摘要 | 中藥炮製的目的是為了減毒及增強療效,所以建立炮製的標準化流程,是建構中藥用安全環境重要的主題之一。然而,杜仲(Eucommia ulmoides Oliv.)之乾燥樹皮為常用滋補肝腎、強壯筋骨,但一般於方劑之中,多用炒制(炒焦、炒黑),少用未炒製之杜仲。因為傳統醫藥典籍記載,炒製後可使其絲斷裂易於切製及萃取率增加,繼而增強功效。然而,炮製的過程,推測其成分應該有了改變,而其主成分為木質類及黃酮類,且松脂醇二葡萄苷(( +) -Pinoresinol di-O-β-D-glucopyranoside)及綠原酸(Chlorogenic acid)為指標成分。因此本計畫將以此兩個成分為主,檢測炒製前後的成分變化,並用 LC-Mass觀察指紋圖譜的消長。目前已收集臺灣市售品 71件中發現,未炒製之杜仲樣品內含有松脂醇二葡萄苷 +)-Pinoresinol di-O-β-D-glucopyranoside)及綠原酸(Chlorogenic acid)的量比炒製品高。實驗室自行炮製結果發現:溫度越高,時間越長則松脂醇二葡萄苷及綠原酸含量越低,但總多酚含量卻增高。炒製過程中噴水或鹽水,則指標成分含量下降程度較少,但與鹽度無成正相關。利用 LPS刺激人類軟骨細胞及兔子膝關節產生發炎現象之試驗模式發現:生品潤製 4小時,切片後 40℃下烘乾,炒製前以(100:35)清水濕潤 10分鐘,180℃下滾筒翻炒 1小時之樣品,指標成分下降較少,總多酚含量提高,且抗關節炎作用最強。但將方法應用至中藥炮製工廠進行,結果不如試驗室炮製理想。推測實驗室建立之杜仲炮製方法,未來至工廠應用時,仍需依設備做修正。 |
英文摘要 | Eucommiae Cortex(Eucommia ulmoides Oliv.) is one of the tonic Chinese herbs. It has been used to strengthen bone function and enhance vital energy. However, the processed Eucommiae Cortex is more popular used than raw material. Because, the processed-Eucommiae Cortex can enhance the pharmacological functions, the bioactive compounds will be changed. In the present investigation, we processed Eucommiae Cortex by ourselves. The quantitative analysis of (+)-Pinoresinol di-O-β-D-glucopyranoside and Chlorogenic acid were detected by HPLC system in raw and processed. The 71 kinds of Eucommiae Cortex were collected from Taiwan markets and analyzed the amounts of the substance markers. The results showing, raw Eucommiae Cortex is richer in (+)-Pinoresinol di-O-β-D-glucopyranoside and chlorogenic acid than processed. In according to our laboratory processed samples of Eucommiae Cortex, the substance markers contain were decrease dependence on temperature and time and not directly the concentration of salty water. However, the total polyphenol contains of processed Eucommiae Cortex were richer than raw. The other hand, the anti-arthritis activity assay of Eucommiae Cortex were evaluated between raw and processed by lipopolysaccharide (LPS) induced inflammation in Primary human chondrocytes and knee of rabbit. In according to bio-assay results, we regulated the best-processed way of Eucommiae Cortex: raw sample was soaked in water for 4 hour, cut into a square and dried under 40℃, soaked in water (100:35) for 10 min before sired-fry and sired-fry for 1 hour under 180 ℃. The method was apple to phaemaceutical factory and the processed products were not well as the Lab. Processed samples. We suggested the processed method of Eucommiae Cortex should be modified by pharmaceutical factory in the future. |
