中文摘要 | 通天草(Uraria crinita)為豆科中草用藥植物,亦稱狗尾苔,零星栽培於台灣中南部。2014年夏屏東地區發現葉片呈現嵌紋、黃化等病毒感染之病株。電子顯微鏡鏡檢,可觀察到長度約800 nm的長絲狀病毒顆粒,罹病細胞內可觀察到風車狀之病毒內含體。機械接種於奎藜(Chenopodium quinoa)進行病毒之單離,9-10日後可觀察葉脈處出現壞疽病斑。單離的病毒分離株回接通天草並未引起感染。萃取通天草罹病葉組織總量RNA,配合Potyvirus屬病毒簡併式引子對進行反轉錄聚合酶連鎖反應(RT-PCR)所增幅出的cDNA片段,其序列與大豆嵌紋病毒(Soybeanmosaic virus, SMV)和Watermelon mosaic virus(WMV)最為相似。解析病毒之全長度基因序列為9651個核苷酸,對應產生一個聚合蛋白。病毒之基因結構與Potyvirus屬病毒相同。病毒全長度基因核苷酸序列與WMV 和SMV的相似度分別為67.6%和70.5%;外鞘蛋白基因核苷酸序列與WMV和SMV的相似度分別為74.0%和74.7%。通天草嵌紋病相關的病毒,應是Potyvirus屬病毒的一個新病毒種。本文是本病毒與本病害的初次紀錄。 |
英文摘要 | Uraria crinita is a Fabaceous plant being used as a medicinal herb and have been sporadically grown in Taiwan. Diseased Uraria leaf samples with symptoms of mosaic, mottle, and yellowing were received from Pingtung of southern Taiwan in the summer of 2014. Filamentous virus-like particles and pinwheel inclusion bodies induced by Potyvirus can be observed in the preparations of Uraria samples under electron microscopic examination. A virus isolate was obtained through mechanical inoculation onto Chenopodium quinoa. However, back inoculation of the virus to Uraria seedlings failed to show symptoms of infection. With Potyvirus degenerate primers and RT-PCR, expected cDNA fragments can be amplified and their nucleotide and amino acid sequences were most similar to that of Soybean mosaic virus (SMV) and Watermelon mosaic virus (WMV). Full-length genome which consists of 9651 nucleotides has been sequences. The genome encodes a polyprotein and its structure is identical to that of Potyvirus. The studied virus is most close to SMV and WMV among Potyvirus members based on their nucleotide sequences of fulllength genome and coat protein (CP) gene. Its full-length genome shares 67.6% and 70.5% similarities to that of SMV and WMV, and its CP gene shares 74.0% and 74.7% to that of SMV and WMV, respectively. Took together, the virus associated to the mosaic disease of Uraria crinita is a tentative new member of Potyvirus. This is the first record of the virus and the disease in Taiwan. |
