中文摘要 | 葉片具有明顯的嵌紋病徵的矮牽牛(petunia, Petunia x hybrid)病株採集自高雄市國立高雄師範大學校園。經三次單斑分離後,取得一病毒分離株,並可回接原寄主(矮牽牛)引起相同病徵。該病毒之型態為球形顆粒,直徑約28-30 nm。寄主範圍測定發現該病毒分離株可感染奎藜、紅藜、菸草、非洲鳳仙花、鳳仙花、茄子、千日紅、白莧菜、雞冠花、菠菜等植物。病毒的耐稀釋度為10-3 倍,熱不活性為75℃,耐保存性為8 天。SDS-PAGE 顯示病毒外鞘由單一種蛋白次單位組成,分子量約為24-25 kDa。以純化的病毒製備同源抗血清,可用於雙向免疫擴散、ELISA 和西方轉漬反應以偵檢病毒。選殖病毒鞘蛋白基因並進行序列解析,得知病毒外鞘蛋白基因(accession number DQ004557)具657 個核苷酸,可對應轉譯218 個胺基酸(分子量為24.1 kDa)。外鞘蛋白基因核苷酸序列經比對,發現其與胡瓜嵌紋病毒II B 亞群最為近似,而與本國的胡瓜嵌紋病毒分離株CMV-Del(AJ131626)、CMV-Eus(AJ131627)、CMV-Ama(AJ131625)等有96%以上的相同度,顯示該病毒為胡瓜嵌紋病毒的矮牽牛分離株(CMV-pet)。本研究是國內首次從矮牽牛分離出胡瓜嵌紋病毒的報告。 |
英文摘要 | Petunia plants (Petunia hybrida) with symptoms of mosaic were found in the campus of National Kauhsiung Normal University. A virus culture was isolated through successive single lesion isolation. The virus infected petunia and produced mosaic symptoms when it was back-inoculated to healthy petunia plants. Spherical virus particles with a diameter of about 28-30 nm can be observed by transmission electron microscopy. The virus isolate infects host plants such as Petunia hybrida. Chenopodium amaranticolor, C. quinoa, Impatiens wallerana, I. balsamina, Nicotiana benthamiana, N. rustica, N. tabacam, Gomphrena globosa, Solanum melongena, Amaranthus mangostanus, Celosia argentea, and Spinacia oleracea. Physical properties of dilution end point (DEP), thermal inactive point (TIP), and longevity in vitro, LIV) were determined as 10-3, 75℃, and 8days, respectively. A single coat protein subunit with a relative molecular weight of about 24-25 kDa was observed in SDS-PAG. The virus was purified and its homologous antiserum was produced. The antiserum detected the virus positively in the double diffusion, ELISA and western blotting tests. The viral coat protein gene was cloned and sequenced (accession number DQ004557). It consists of 657nucleotides and encodes a coat protein with molecular weight of 24.1 kDa。The nucleotide sequence of studied coat protein gene was highly homologous to that of subgroup IB isolates of Cucumber mosaic virus (CMV) isolates, specially to some domestic CMV isolates such as CMV-Del (AJ131626), CMV-Eus (AJ131627), and CMVAma(AJ131625). This is the first study of the natural CMV infection to petunia in Taiwan. |
