中文摘要 | 孤挺花潛隱病毒(Hippeastrum latent virus, HiLV)是由輕微嵌紋病徵的孤挺花葉片的粗汁液以機械方式接種於奎藜上,再經奎藜三次單分離純化而得到的。病毒粒子呈長絲狀,長度約為626 nm。孤挺花潛隱病毒在分類上是屬於Carlavirus。利用cDNA 合成技術及PCR 技術,增幅孤挺花潛隱病毒基因組片段來分析孤挺花潛隱病毒的基因序列。孤挺花潛隱病毒的全長基因組有8500 個核苷酸,不包括poly A tail(accession number: DQ098905),其鹼基組成為2281 個A(26.83%)、1722 個C(20.25%)、2274 個G(26.75%)和2223 個U(26.15%)。全長基因組RNA 有6 個開放轉譯架構,從5'到3'端依序可產生的蛋白質為223.5 kDa(1971 aa)、25.5 kDa(230 aa)、11.6kDa(108 aa)、6.6 kDa(59 aa)、32.5 kDa(292 aa)和12.1 kDa(105 aa),分別為RdRp、TGB 1、TGB 2、TGB3、CP 和NABP 基因。其中TGB 1 和TGB 2 基因有重疊,TGB 2 和TGB 3 基因有重疊,CP 和NABP 基因有重疊,這些皆為典型的carlaviruses 之特性。在各蛋白質皆可發現有保留的motif 的胺基酸序列,而特定的核苷酸序列,除了有次基因組RNA 的病毒會有的核苷酸序列和polyadenylation signal 的核苷酸序列沒有之外,其餘皆有。將所完成的HiLV 全長度核苷酸與啤酒花潛隱病毒(Hop latent virus, HpLV)、百合無症病毒(Lily symptomless virus, LSV)、楊樹花嵌紋病毒(Poplar mosaic virus, PopMV)和馬鈴薯病毒(Potato virus M, PVM)等4 個病毒作各基因的核苷酸序列相同度及胺基酸序列相似度,比較結果都偏低(20~45%)。在Genbank 上也沒有與HiLV的核酸序列有相近的植物病毒,就目前而言,HiLV 沒有與之同源性高的植物病毒。 |
英文摘要 | Hippeastrum latent virus (HiLV) was originally isolated from symptomless or mosaic symptom of Hippeastrum hybridum in Taiwan. HiLV caused local lesions on Chenopodium quinoa, C. amaranticolor and Gomphrena globosa. HiLV isolated from local lesions of Chenopodium quinoa inoculated mechanically from symptomless or mild mosaic symptom amaryllis leaves. HiLV were purified from C. quinoa. HiLV is a member of Carlavirus genus with the length of 626 nm and the size of coat protein was around 35.4 kDa. cDNA of HiLV was amplified by PCR and sequenced, revealing that the full length genome RNA of HiLV contains 8500 nucleotides, excluding the poly(A) tail. The genomic RNA of HiLV contains six open reading frames (ORFs) encoding RNA-dependent RNA polymerase (RdRp), triple gene block (TGB) 1, TGB 2, TGB 3, coat protein (CP) and nucleic acid binding protein (NABP). The coding sequences between TGB 1 and TGB 2 genes, TGB 2 and TGB 3 genes and CP and NABP genes are overlapping, phenomenon commonly found in carlaviruses. Conserved amino acid and nucleotide sequences commonly found in carlaviruses also were present in HiLV. Comparison of nucleotide and amino acid sequences revealed that HiLV was not closely related to HpLV, LSV, PopMV and PVM, showing low identities and similarities (20~45%). Thus, HiLV is likely a novel virus. |
