中文摘要 | 病毒病害對台灣的甘藷栽培產業是一大威脅,在田間多呈現病毒複合感染的狀態,使得甘藷病徵更形嚴重外,且會造成甘藷減產。本研究選出10種甘藷栽培種(Ipomoea batatas),包括台農57、64、66、67、68、69、70、71、72及73號等,以及10種組織培養保存之野生種甘藷(Ipomoea trifida)(T1, T2, T3, T4, T5, T6, IT7-1, IT8-1, PI-1和PI-2),針對台灣常見的3種馬鈴薯Y病毒屬(Potyvirus)的病毒,包括甘藷羽狀斑駁病毒(Sweet potato feathery mottle virus, SPFMV)、甘藷潛伏病毒(Sweet potato latent virus, SPLV)及甘藷G病毒(Sweet potato virus G, SPVG),利用嫁接接種的方式分析甘藷對病毒的抗性,觀察病徵並以反轉錄聚合酶連鎖反應(reverse transcription-polymerase chain reaction, RT-PCR)確認病毒之感染與否,以找出可能存在的抗病品系。10個台農栽培種甘藷在接種三種病毒30-40天後,除了台農68外,其餘9個品種皆出現嵌紋、斑駁、葉部扭曲等病徵,且以RT-PCR檢測出這9個品種已受上述1-3種病毒之感染。而台農68甘藷雖病徵不明顯,但檢測到SPFMV及SPVG之感染。10種野生種甘藷在嫁接接種後50-60天仍未呈現明顯而典型的病毒感染病徵,經RT-PCR分析結果顯示10種野生種甘藷對三種病毒均呈陰性反應,顯示野生種甘藷中可能存在有天然的抗病基因,可當做抗病毒病害育種之材料,並做為培育健康種苗之參考。 |
英文摘要 | Virus disease is the main threat to sweet potato production in Taiwan. Mixed infections of viruses commonly occurred in cultivated fields of sweet potato, and caused severe symptoms and yield reduction. In this study, ten sweet potato (Ipomoea batatas) Tainung varieties (TN 57, TN64, TN66, TN67, TN68, TN69, TN70, TN71, TN72 and TN73) and ten wild type sweet potato lines (Ipomoea trifida) (T1, T2, T3, T4, T5, T6, IT7-1, IT8-1, PI-1 and PI-2) were assessed for resistance to Sweet potato feathery mottle virus (SPFMV), Sweet potato latent virus (SPLV) and Sweet potato virus G (SPVG) by a graft-inoculation assay. Observation of symptom development and reverse transcription-polymerase chain reaction (RT-PCR) assay were performed to demonstrate virus infection. Nine Tainung varieties, except TN68, showed obvious symptoms including mottle, mosaic and distortion of leaves 30-40 days after inoculation, and the virus infections were verified by RT-PCR. No obvious symptom was observed on TN68, but RT-PCR result showed that SPFMV and SPVG infections occurred. All of ten wild type sweet potato lines showed no obvious symptom 50-60 days after graft-inoculation and virus negative in RT-PCR assay. Our results suggest that wild type sweet potato lines probably carry natural resistance genes to the inoculated potyviruses and could be used as the materials for breeding. |
