中文摘要 | 本研究在分析不同培養條件,包括pH、溫度及硝酸鈉對螺旋藻藻藍素產量的影響,及以反應曲面法探討其最適產藻藍素之培養條件。實驗利用修飾之BG-11培養基於光度85 μmol m^(-2)s^(-1)及連續打氣條件培養螺旋藻14天後,以凍結與解凍萃交替方法萃取藻藍素。實驗是應用Box-Behnken Design (BBD)設計影響藻藍素產量之三獨立三階層變量因子:[X1 : pH (7、9、11);X2:生長溫度(30、33、36°C); X3:硝酸鈉濃度(1.5、2.25、3 g/l)]。結果顯示,溫度與硝酸鈉濃度顯著地影響藻藍素之產量。高產量藻藍素之最適化培養條件為pH 9、溫度 33.71°C及硝酸鈉濃度3 g/l下,可達最高產量91.09 ± 2.16 mg/g之藻藍素。 |
英文摘要 | This study investigates the influence of independent process variables, including pH, temperature and sodium nitrate on the C-phycoyanin (C-PC) yield of Spirulina platensis. Modified BG-11 medium was used for growing S. platensis for 14 days under 85 μmol photon m^(-2) s^(-1) light intensity and continuous aeration. Extraction of phycocyanin was done by using freeze-thaw method. A Box–Behnken design (BBD) was used to evaluate the C-PC yield of S. platensis under the influence of three independent variables at three level [X1: pH (7, 9 and 11); X2: growth temperature (30, 33 and 36oC); X3: sodium nitrate concentration (1.5, 2.25 and 3 g/l)]. The results showed that C-PC yield significantly influenced by temperature and sodium nitrate concentration. The optimum condition for highest yield of C-PC (91.09 ± 2.16 mg/g) was obtained at pH 9, temperature 33.71˚C, and sodium nitrate concentration 3 g/l. |
