| 中文摘要 | 本實驗主要目的在研究蛇床子素奈米粒子降血脂之藥效學,希望發展生物可降解性包覆方法,且大幅增加生體可用率的藥物劑型。蛇床子素的奈米粒子以高速均質法製備,包覆的材料為聚乳酸-甘醇酸,檢測包埋率及粒徑大小。在降血脂的藥效學方面,以敘利亞倉鼠為動物模式,為期八週的實驗,分為高脂肪食物、高脂肪食物合併蛇床子素奈米粒子及高脂肪食物合併 fenofibrate 三組。血液生化檢驗值顯示,fenofibrate 與蛇床子素奈米粒子可有效降低三酸甘油酯及總膽固醇;整體降血脂成效而言,fenofibrate 與蛇床子素奈米粒子幾乎不相上下。肝臟病理切片方面,fenofibrate 與蛇床子素奈米粒子可有效改善脂肪肝狀態。 |
| 英文摘要 | The objective of this investigation was to compare hypolipidemic performances between osthole nanoparticles and fenofibrate as well as to develop a biodegradable polymeric drug delivery system for osthole. Osthole nanoparticles loaded poly (lactic-co-glycolic acid) was prepared by high speed homogenization. In vitro physicochemical characterizations, such as encapsulation efficiency and particle size test were estimated. With regard to pharmacodynamic examinations, Syrian hamsters (Mesocricetus auratus), which were used as an animal model, and simultaneously administered orally 20 mg/kg of osthole nanoparticles and fenofibrate once daily for eight weeks. Blood biochemical data of experimental hamsters showed that osthole nanoparticles and fenofibrate evidently reduced triglyceride and total cholesterol in contrast with the high fat diet-fed group. In vivo hypolipidemic effectiveness of these controls were ranked as follows, osthole nanoparticles ≒ fenofibrate. Histological examinations also uncovered that both osthole nanoparticles and fenofibrate group providing superior defense to alterations of fatty liver in contrast with the high fat diet group. |
