中文摘要 | 水稻徒長病為水稻古老之病害,由病原真菌Fusarium fujikuroi所引起,在台灣已成為風土病,每年都會發生,造成輕、重不等之危害。種子消毒為主要之防治方式,然而此病害仍舊不定期在田間發生。抗病育種是病害防治之重要策略,然而,其前提是必須知道台灣水稻栽植品種之抗感性,本實驗之目的即在建立一套標準流程,運用於大量篩檢水稻品種對徒長病菌之抗感性。選用高毒力之菌株IL01,在菌株最適生長溫度28°C下,以7個水稻品種(台稉9號、台南11號、台稉16號、台東30號、台農71號、高雄139號、台中秈10號),用不同孢子濃度,接種催芽後之水稻種子,結果顯示10^3 spores/mL之接種濃度,最能夠使水稻呈現出不同之抗感性差異,可作為篩檢水稻抗感性品種之病原菌接種濃度,而評估抗感性則以各品種之罹病率為指標。 |
英文摘要 | Bakanae disease, caused by Fusariumfujikuroi, is one of the oldest rice diseases and is endemic in Taiwan for years. Seed sterilization is the major approach to control this disease. However, this disease still outbreaks from time to time. In order to establish a sustainable control method, breeding resistant rice variety is always the major concern. The information regarding susceptibility of rice varieties to the pathogen of bakanae disease is therefore essential. The aim of this study was to standardize the screening method to evaluate the susceptibility of rice varieties. A high virulent isolate, IL01, was selected to be the source of inoculum. Optimal temperature for IL01 to grow is 28°C. Seven varieties, Taikeng 9, Taichung sen 10, Tainan 11, Taikeng 16, Taitung 30, Tainung 71, Kaoshiung 139 were tested. Germinated seeds were inoculated with different concentrations of spore suspension, and were then planted on Akadama soil on seedling trays. Disease incidences were recorded 21 days after inoculation. It revealed that inoculum concentration of 10^3 spores/mL was suitable for discriminating susceptibilities of rice varieties. A standardized procedure for determining susceptibility has been established. High virulent isolate IL01 in the concentration of 103 spores/mL is inoculated to germinated seeds, which is incubated at 28°C for 21 days. Susceptibility is evaluated by disease incidence. |
