中文摘要 | 本研究分別以魚腥草、白鶴靈芝、玉米鬚和金錢草藥渣取代木屑的不同配方培養基,探討栽培松杉靈芝和雲芝的可行性,同時亦探討靈芝和雲芝子實體之多醣體和粗三萜類的含量及抗氧化能力的分析。將松杉靈芝和雲芝菌種分別接種到不同比例添加白鶴靈芝、魚腥草、金錢草和玉米鬚等藥渣的木屑混合培養基,置於28℃的培養室,不照光下進行菌絲培養和子實體出菇試驗,分別記錄上述不同培養基配方的菌絲生長速度和子實體產量。將不同配方培養基生產的子實體進行測定多醣體和粗三萜類的含量,同時以甲醇萃取子實體,其萃取液分別進行清除DPPH 自由基能力、螯合亞鐵離子能力和還原力分析。由子實體產量、生物效率和多醣體含量的結果顯示,以玉米鬚為代料的培養基最適合栽培松杉靈芝和雲芝。若以粗三萜類含量的結果顯示,則以金錢草取代2/3 木屑和完全取代木屑的培養基最適合栽培松杉靈芝和雲芝。在抗氧化能力方面,以魚腥草和金錢草藥渣的培養基生產之松杉靈芝和雲芝子實體甲醇萃取液,清除DPPH自由基能力較玉米鬚藥渣的培養基生產之松杉靈芝和雲芝高;在螯合亞鐵離子能力方面,魚腥草、玉米鬚和金錢草藥渣的培養基生產之松杉靈芝和雲芝子實體甲醇萃取液均低於對照組;在還原力方面,松杉靈芝和雲芝子實體甲醇萃取液濃度達20mg/ml時,均略低於對照組。 |
英文摘要 | The aim of this study was to partially replace sawdust by four herbal dregs (Houttuynia cordata, Rhinacanthus nasutus, Maydis stigma and Desmodium styracifolium) for Ganoderma tsugae and Coriolus versicolor cultivation, respectively; meanwhile, to explore the mycelial growth rates and fruiting body yields of these two medicinal mushrooms on various media and to measure the polysaccharide and triterpene contents of their fruiting bodies, and to analysis antioxidant activities of methanol extracts from their fruiting bodies. The various combination substrates were filled into polyethylene bags and sterilized at 121℃ for 60 min. After cooling the substrates down to room temperature, then they were inoculated with the grain spawn. The inoculated substrates were kept in a spawn running room at 28℃ under dark condition. The mycelial growth rates and fruiting body products in various substrates for Ganoderma tsugae and Coriolus versicolor were record. The polysaccharide and triterpene contents of fruiting bodies were measured by phenol-sulfuric acid method and organic solvent extract method, respectively. The scavenging ability on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, chelating ability on ferrous ions and reducing power were measure by spectrophotometer method, respectively. From the results of fruiting body yields, biological efficiency and polysaccharide content, the optimal medium for Ganoderma tsugae and Coriolus versicolor cultivation were the medium partially replace sawdust by Maydis stigma. And From the result of triterpene content, the optimal medium for Ganoderma tsugae and Coriolus versicolor cultivation were the medium replace 2/3 sawdust and complete substitution by Desmodium styracifolium. The scavenging ability on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals of methanol extracts from Ganoderma tsugae and Coriolus versicolor fruiting bodies cultivated on the substrates replace by Houttuynia cordata and Desmodium styracifolium were higher than those of fruiting bodies cultivated on the substrates replace by Maydis stigma. The chelating ability on ferrous ions of methanol extracts from Ganoderma tsugae and Coriolus versicolor fruiting bodies cultivated on Houttuynia cordata, Maydis stigma and Desmodium styracifolium were lower than that of control. The reducing power of methanol extracts from Ganoderma tsugae and Coriolus versicolor fruiting bodies cultivated on the substrates replace by these three herbal dregs were slightly lower than that of control at 20 mg/ml. |
