| 中文摘要 | 本研究探討水稻根缺鐵時對於鎘的吸收及OsIRT1 基因表現之影響。水稻幼苗根缺鐵時會增加鎘吸收外,亦會提高OsIRT1基因的表現。使用NO專一性螢光探針偵測水稻根中內生NO時,發現缺鐵時會誘導螢光增加。若缺鐵時處理 NO清除劑cPTIO,除了降低水稻根中螢光表現外,也同時會減少鎘的吸收與OsIRT1基因的表現。由以上結果可知,NO會參與水稻根缺鐵時誘導鎘之累積與提高OsIRT1基因之表現。 |
| 英文摘要 | The effects of iron (Fe) deficiency on the uptake of cadmium (Cd) and expression of OsIRT1 in rice roots were examined. Roots of Fe-deficient seedlings had higher Cd accumulation than those of Fe-sufficient seedlings. Iron deficiency also enhanced the expression of OsIRT1 gene in rice roots. Endogenous nitric oxide (NO) was detected by the specific fluorescent probe 4-amino-5-methylamino-2', 7'- difluorescein diacetate. Iron deficiency was effective in inducing NO fluorescence, which was blocked by the specific NO scavenger 2- (carboxyphenyl)- 4,4,5,5- tetramethylimidazoline -1-oxyl-3-oxide (cPTIO). The scavenging of NO by cPTIO prevented Fe deficiency-enhanced uptake of Cd and expression of OsIRT1 was also observed. Results suggest that NO is required for Fe deficiency-induced Cd accumulation and OsIRT1 expression in rice roots. |
