中文摘要 | 本研究取用虎杖(Polygonum cuspidatumSieb. et Zucc.)無菌播種苗之葉片作為培殖體,經農桿根群菌(Agrobacterium rhizogenes)BCRC 15011感染後移置於1/2 MS基礎培養基上,在誘導出毛狀根(hairy root)後,以高效液相層析法(high performance liquidchromatography;HPLC)進行白藜蘆醇(trans-resveratrol;t-Res)含量分析。毛狀根編號1-11及1-13二根群於MS培養基中生長緩慢,但以液態培養於1/2 MS或B5培養基中,二者生長量均顯著增加且優於1/2 B5液態培養基(p < 0.05)。三種液態培養基中又以1/2 MS所培養的毛狀根內所含的t-Res最高。在誘引因子的試驗中以紫外線(ultraviolet;UV)照射處理後之毛狀根,t-Res的含量可提升至原來的兩倍。在幾丁聚醣(chitosan)的誘引試驗中,以10 mg L^-1chitosan添加在1/2 MS液體培養基內並黑暗培養48 h,毛狀根1-11根群內t-Res的含量可提高到26.62±0.85 μg(g DW)^-1,約為未處理的四倍。 |
英文摘要 | Detached leaves of in vitro grown Polygonum cuspidatum Sieb. et Zucc. seedlings were used to induce hairy roots by subculturing on 1/2 MS medium after infection of Agrobacterium rhizogenes (strain BCRC 15011). High performance liquid chromatography (HPLC) was adopted to analyze the content of trans-resveratrol (t-Res) in the root cultures. Two hairy-root lines (1-11 and 1-13) exhibited slow growth in MS medium, but had a better growth (biomass increase) in liquid form of 1/2 MS and B5 media (p < 0.05), both superior than in 1/2 B5 liquid medium. When incubated in 1/2 MS liquid medium, root cultures produced more t-Res. By ultraviolet (UV) illumination treatment, the content of t-Res was doubled in the treated roots. When incubated in the dark with addition of 10 mg L^-1 chitosan in 1/2 MS liquid medium, the content of t-Res was found increased to 4-fold [6.62 ± 0.85 μg (g DW)^-1] in hairy-root line 1-11. |
