中文摘要 | 含有CpG模組的寡核苷酸(oligodeoxynucleotides;ODN)具有種別專一性,運用於人、豬及牛羊等哺乳動物用疫苗研究,可以增強疫苗之免疫效能。本研究以雞隻周邊血液單核球細胞(peripheral bloodmononuclear cells;PBMC)和脾臟細胞(splenocyte)為生體外篩選系統,以促進interferon-γ(IFN-γ)基因表現為指標,篩選出於禽類具有佐劑效能之CpG ODN-D2,並以此CpG ODN分別搭配雞新城病(Newcastle Disease;ND)之活毒疫苗/ND不活化疫苗,或禽流感(Avian Influenza;AI)不活化疫苗免疫雞隻,實驗結果顯示以CpG ODN-D2作為疫苗佐劑,可有效增加抗ND或AI之特異性血球凝集抑制抗體、顯著促進脾臟細胞之增殖作用;以CpG ODN-D2搭配家禽霍亂不活化菌苗免疫小鼠,可有效增加攻毒後之存活率。因此,本實驗成功建立以生體外篩選系統,評估禽類專一性CpG ODN佐劑之免疫活性,CpG ODN-D2可運用於多種家禽疫苗,以增加其疫苗有效性。 |
英文摘要 | Oligodexoxynucleotides (ODN) with species-specific CpG motifs can modulate the immune functions and have been used as an effective adjuvant in human or other mammalian vaccines. In this study, chicken peripheral blood mononuclear cells (PBMC) and splenocytes were used as the in vitro screening systems to screen the CpG ODNs which effectively increased IFN-γ gene expression. Then, the avian-specific ODN-D2 was applied as an adjuvant in the Newcastle disease (ND) live/inactivated vaccine or avian influenza inactivated vaccine in SPF chicken. Resuts showed that ODN-D2 as an adjuvant significantly increased the HI titers against ND or AI. In addition, cell proliferation of splenocytes was also enhanced in those inoculated with ODN-D2 co-administrated vaccines. Furthermore, the highest survival rate was observed in mice vaccinated with fowl cholera inactivated bacterin (FC) co-administrated with CpG ODN-D2. These results indicated that the in vitro systems can screen the effecti ve avian-specific CpG ODN and the resultant CpG ODN-D2 may provide the valuable immunoadjuvant efficacy in chicken vaccines. |
