中文摘要 | 摘要 本研究以水稻秈稉雜交分離後代為材 料,利用逢機擴增多型性 DNA ( random amplified polymorphic DNA; RAPD )分子 標誌技術來建立巨胚及非巨胚稻之DNA 具 多型性條帶,並進行巨胚稻及非巨胚稻發芽 前後總胺基酸含量分析,以選育巨胚水稻新 品系。試驗結果自34 組逢機引子(random primer)中篩選出9 組可有效判別巨胚及非巨 胚稻親本,共擴增出19 條具差異性之DNA 片段,除協助應用於雜交分離後代之判別 外,進一步更將純化這些差異性條帶並定 序,了解其於基因組中之定位,再據此重新 設計穩定且可信賴之引子,作為判別巨胚稻 與非巨胚稻之分子標誌。此外,由總胺基酸 含量分析結果顯示, 巨胚稻之 γ-aminobutyric acid (GABA)含量明顯高於 非巨胚稻,處理成發芽米後,兩者GABA 含 量差異更為顯著,值得進一步選育成為機能 性水稻新品系。 |
英文摘要 | ABSTRACT The objective of this study was to use random amplified polymorphic DNA (RAPD) analysis to assist the breeding of new rice variety with giant embryo. The total amino acid content in brown rice with/without germination treatment was also determined to clarify the relationship between giant embryo and γ-aminobutyric acid (GABA) content. The segregation lines selected from aromatic Lemont (indica type rice) and NKY203 (japonica type rice) intercross were used. At first, 34 sets of random primers were chosen for polymerase chain reaction. It showed that 9 sets of random primer could amplify 19 amplicons in total with lengths from 450 bp to 2.8 kb, showing good resolution to discriminate giant embryo and non-giant embryo rice. These differentiated amplicons can be used to sequencing and re-designing stable primer to become reliable DNA markers related to giant embryo in the follow-up experiments. Besides, the results showed that GABA content in giant embryo rice is significantly higher than non-giant embryo rice, especially after germination treatment. In conclusion, several potentially useful DNA markers were derived from RAPD analysis that may be used to assist the selection of rice new variety with giant embryo. |
