中文摘要 | 本研究旨在使用中醫藥物-骨碎補結合低強度超音波(Low Intensity Pulsed Ultrasound, LIPUS),針對類造骨細胞MG-63細胞所做的結合刺激研究,並利用可測定細胞存活率的MTT assay(3-(4, 5-dimethyl thazol-2-yl)-2, 5-diphenyl-tetrazolium bromide)技術,來觀察超音波與骨碎補的存活反應,以及偵測造骨作用的生化指標-鹼性磷酸酶鹼性磷酸酶(Alkaline Pphosphatase, ALP)活性,藉此,觀察其結合刺激效應對造骨作用的效用。經實驗結果得知,骨碎補分別以濃度10、20、50、100、300μg/ml加入實驗組細胞中,顯示骨碎補在濃度50μg/ml作用時,和控制組(100%)相比,有顯著的存活反應(155.91%),顯示骨碎補對MG-63而言,具有細胞增殖效應,而單獨使用頻率1MHz強度0.05、0.15以及0.3W/平方公分之LIPUS刺激MG-63細胞3min時,在強度0.05W/平方公分時可發現,有刺激細胞增殖效應(112.88%)。而結合兩種方法來刺激類骨細胞MG-63,結果顯示,0.15W/平方公分結合骨碎補50ug/mL有最佳的細胞存活反應(121.66%),而將兩者不同的刺激結合並觀察其促進鈣化機制的ALP活性表現可發現,骨碎補結合LIPUS強度0.15W/平方公分時,和只使用骨碎補刺激的組別相比,有最佳的ALP活性(105.23%),因此,骨碎補結台LIPUS對存活率以及ALP活性,皆有刺激上升的效果。 |
英文摘要 | It has been suggested that Chinese herbal medicine Rhizoma Drynariae or low-intensity pulsed ultrasound (LIPUS) can accelerate bone mineralization and regeneration. However, the effect of Rhizoma Drynariaea combined LIPUS on the above description is unclear. The purpose of our research project is to determine if Rhizoma Drynariaea combines LIPUS treatment can have effects on cell viability and alkaline phosphatase activity in an osteoblast like cell line. A human osteoblastic cell line, MG-63, was analyzed in different concentrations (10、20、50、100、300μg/mL) of the Rhizoma Drynariaea and the best concentration on MG-63 cell viability was 50μg/mL. Next, we cultured MG-63 cells in the optimal concentration of Rhizoma Drynariaea and treated with different LIPUS intensities (50、150、300 mW/cm^2). After 48 hours incubation, cells were collected and analyzed their viability and ALP activity by MTT assay and ALP test kit. Results showed when Rhizoma Drynariaea (50μg/mL) combined LIPUS in 0.15 W/cm^2 intensity could efficiently promote both the cell proliferation (121.66%) and ALP activity (105.23%) of MG-63 cells. Therefore, we suggest that Rhizoma Drynariaea and LIPUS have synergistic effect on proliferation and mineralization of human osteoblastic cells. |
