中文摘要 | 鳥糞嘌呤(guanine)是去氧核醣核酸(DNA)中最能表現出電化學特性的成分,而去氧核醣核酸又常會因一些土壤微生物或病毒的解體游離到土壤溶液中。因此,若能以迅速方便的方法來偵測鳥糞嘌呤,則對未來在土壤微生物或病毒釋出去氧核醣核酸方面的探討應具有發展潛勢。以三種不同種類黏土礦物先修飾在環盤電極上,再覆蓋含有三聯吡啶釕(Ru(bpy)3(上标 2+)之全氟磺酸聚合物(Nafion)薄膜,對鳥糞嘌呤之偵測均可獲得不錯的穩定度,且其對0.05~1.00 mM之鳥糞嘌呤的測定結果也有良好的測定關係(R^2=0.986~0.996)。三種黏粒修飾電極中,以KGa-1N1R2修飾電極所測得之氧化電流量最大,此電極搭配流動注入系統對標準品DNA偵測,其測得之59.12 μM鳥糞嘌呤與標準品中65.69 μM鳥糞嘌呤比較,顯是有90%準確性,以此修飾電極快速方便偵測鳥糞嘌呤應有其發展潛勢。 |
英文摘要 | Guanine is an electroactive component of deoxyribonucleic acid and soil microorganisms or viruses can be destroyed to release deoxyribonucleic acid. Therefore, it is important to develop a rapid and convenient method to study released deoxyribonucleic acid of soil microorganisms and viruses. In this study we have used three clay modified electrodes modified with Nafion and Ru (bpy) 3(superscript 2+) to determine guanine stability. A linear calibration curve (R^2=0.986~0.996) was obtained in the range of 0.05~1.00 mM guanine. The KGa-1N1R2 modified electrode showed the largest oxidative current among the three clay modified electrodes studied. Most importantly, ~90% of accuracy was achieved in the determination of a standard DNA containing 65.69 μM guanine using the KGa-1N1R2 modified electrode by flow injection analysis. Overall, this clay modified electrode possesses high potential for the screening of guanine in soil microorganisms or viruses. |
