中文摘要 | 豬假性狂犬病病毒(pseudorabies virus, PRV)被認為可能造成宿主之免疫抑制。樹狀突細胞(dendriticcell, DCs)在傳染病之免疫反應中扮演啟始、調節及維持之重要角色。本試驗擬探討豬隻骨髓細胞衍生之未成熟樹狀突細胞(bone marrow-derived immature dendriticcells, BM-imDCs)對PRV之感受性,以及BM-imDCs受PRV感染後其細胞表面抗原及功能的改變。結果顯示,PRV能感染豬隻BM-imDCs且能有效的繁殖於細胞內。PRV感染造成BM-imDCs細胞表面分子的表現下降,亦對細胞之吞噬功能及分泌IL-10與IL-12的能力造成抑制。此免疫抑制作用可能在豬隻受PRV感染後之二次性病原感染上扮演重要角色。PRV感染後造成BM-imDCs細胞表面抗原及功能改變的因子及其機序仍待進一步探討。 |
英文摘要 | Pseudorabies virus (PRV) infection has been suggested to induce an immunosuppressive effect on the host. Dendritic cells (DCs) play the critical role in the initiation, regulation and maintenance of immune responses against infections. In this study, we investigated the phenotypic and functional alterations of porcine bone marrow-derived immature DCs (BM-imDCs) after the infection of PRy. Results showed that PRV can infect porcine BM-imDCs with successful replication in cells. PRV infection impaired phenotypic expression, phagocytosis and the LPS-induced [-10 and IL-12 production by BM-imDCs. These effects may explain the potential role of PRV in the induction of immunosuppressive effect and predisposing infected pigs to secondary infection. The mediators and the mechanisms by which the PRV infection down-regulates the phenotypic expression and funct ion in porcine BM-imDCs requires further investigation. |
