中文摘要 | 本試驗目的在探討抗緊迫藥物azaperone對離乳後併欄仔豬下視丘-腦下垂體-腎上腺軸(Hypothalamus-pituitary-adrenal axis, HPA axis)的影響。選用18頭28日齡來自不同6胎之仔豬,每胎3頭,逢機分配至3組:緊迫組(S組)直接併欄,藥物處理組(T組)於併欄時注射azaperone,以及對照組(C組)留在分娩舍。所有仔豬於併欄前及2小時後採血分析皮質醇(cortisol)值。第二次採血後將仔豬以電昏後放血犧牲,迅速取出下視丘、腦下垂體、腎上腺等組織置入液態氮中,並以即時反轉錄聚合酶鏈反應(real-time RTPCR)分別測定下視丘c-fos及CRH、腦下垂體c-jun及CRHR1、以及腎上腺CRHR1及MC2R等基因表現。結果顯示,併欄前後之血漿皮質醇值僅在S組有升高達顯著水準(P<0.05)。各基因表現以C組為基準進行比較,結果為S組及T組下視丘c-fos向下調(downregulation),而腦下垂體CRHR1則呈向上調控(up-regulation),而S組與T組間各基因表現均無顯著差異。由以上結果得知離乳後併欄對仔豬會造成緊迫及HPA軸中下視丘、腦下垂體活性改變,而注射azaperone可以減緩緊迫但不影響HPA活性。 |
英文摘要 | This study was conducted to investigate the effect of azaperone on the plasma cortisol and stress-related gene expression in the hypothalamus-pituitary-adrenal (HPA) axis alteration in co-mingling weaned pigs. Eighteen 28 days old piglets from 6 litters were allocated to 3 groups, S: co-mingled group, T: azaperone treatment group and C: piglets stayed beside their dams in the far- rowing house. Blood samples were collected prior and 2 hours after co-mingling to determine the plasma cortisol change. After the second blood sampling, all animals were electrically euthanized and the HPA tissues were quickly removed for target gene mRNA quantification by real-time RT-PCR. The results showed that the concentrations of cortisol in S group are significantly higher (P<0.05) than those in other groups. The gene expressions of hypothalamic c-fos in S and T group are down- regulated, and expressions of pituitary CRHR1 in S and T groups were up-regulated when compared to that in the C group. No significant differences on all these genes between S and T group were found. Thus, the co-mingling process was stressful to the weaned pigs, and azaperone may reduce the stress without influencing the stress-related gene expressions in the HPA axis. |
