中文摘要 | 定位數量性狀基因座(quantitative trait loci, QTL)的統計方法已被廣泛地討論。然而大部分存在的方法需要表現型為常態分配且可完整觀察的假設,當表現型為事件時間(time-to-event)資料時,常有偏斜分配與失去追蹤等問題,因而違背這些假設。在本文中,以半母數加速失敗時間模式(semiparametric accelerated failure time model; Wei 1992)描述QTL對會有設限失敗時間的表現型之影響。又發展秩有關的推論(rank-based inference)估計迴歸參數,並使用計分檢定(score test)取代傳統的概似比檢定,搜尋染色體上的QTL。另一個定位QTL的難題是在相同基因組上所執行多重檢定,如何決定一適合的門檻值(threshold value)用以宣告QTL的存在。本研究採用Zou et al. 英文摘要 | Statistical methods for mapping quantitative trait loci (QTL) have been widely discussed. While most existing methods assume that the phenotype is normally distributed and completely observed, these assumptions may be violated when the phenotype is time-to-event, which has a skewed distribution and is often low to follow-up. In this article, the effect of QTL on the censored failure-time phenotype was formulated through the semiparametric accelerated failure time model (Wei 1992). The approaches utilizing rank-based inference were developed to estimate regression parameters, and the score tests, instead of traditional likelihood ratio tests, were employed to search the entire chromosome for QTL. An extra challenge for mapping QTL is how to determine an appropriate threshold value for declaring the presence of QTL in multiple tests on the same genome. The resample approach proposed by Zou et al. (2004) was extended to determine the threshold value for multiple tests. Several simulation studies are presented to evaluate the performance of the proposed methods. |
