中文摘要 | 本研究採集陽明山、新屋、三灣、大肚山、八卦山、梅山、關仔嶺、茂林、恆春等地9個族群,共90株單株。分析其簡單序列重覆區間(Inter simple sequence repeat, ISSR)的差異以研究其族群間遺傳變異及基因交流情形,提供族群資源保育與遺傳研究之參考依據。 測試100個UBC隨機序列引子,選取具有多型性及再現性良好的6個引子進行聚合?鍊式反應(Polymerase chain reaction, PCR),獲得74個條帶,多型性條帶有64個(86%)。並經由NTSYS-pc ver. 2.0與POPGENE ver. 1.31套裝軟體分析此ISSR數據。據AMOVA分析所得,受測之群組在遺傳結構上,以族群內變方成分百份比占全部變方成份的83.73%,族群間占16.27%,顯示相思樹的遺傳變異主要存在於族群內。相思樹族群的平均總基因歧異度值(Ht)為0.4512;而族群內(Hs)的平均基因歧異度為0.3666;族群間的平均基因歧異度值(Dst)為0.0846。以POPGENE分析得知基因流傳值(Nm)為2.1664,遺傳結構中的遺傳分化係數(Gst)為0.1875,代表族群間的變異僅占總變異的18.75%。隔離分化檢測表明地理距離和遺傳距離的相關值為正值,呈顯著相關性(r=0.342, p=0.027<0.05)。 |
英文摘要 | In this present report of we collected 9 populations (90 individuals) of Acacia confusa from Yangming Mountain, Sinwu, Sanwan, Dadu Mountain, Bagu Mountain, Meishan, Guanzailing, Maolin and Hengchun forest sites respectively. The genetic variation among these populations was investigated using ISSR (Inter simple sequence repeat) analysis to study the genetic diversity and the circumstances of gene flow among these populations. The results provide useful information for conservation of population resources and also for the related studies in genetics. One hundred UBC random primers were primary examined for obtaining the optimized reaction conditions during the treatment of polymerase chain reaction (PCR) and 6 primers were finally selected. A total amount of 74 bands were produced and 64 of them were polymorphic (86%). All the ISSR information was analyzed using softwares NTSYS (pc ver. 2.0) and POPGENE (ver. 1.31). The AMOVA results revealed that the component of variance among populations is 16.27% and is attributed to the population differentiation. The main variance component was counted at 83.73% due to differentiation within populations; this variance within populations was shown to be highly significant. The populations we studies performed an average value of total genetic diversity (Ht) at 0.4512; the average value of genetic diversity within populations (Hs) is 0.3666 and value of genetic diversity among populations is 0.0846 (Dst). Analysis applied by POPGENE demonstrated that there is a remarkable genetic diversity (Gst=0.1875 and Nm=2.1664) which indicates that the variation among populations was 18.75% of the total variations. Mantel test indicated that there is a positive correlation (r=0.342=0.027<0.05) existing between geographical and genetic distances. |
