中文摘要 | 本研究之目的在於探討免疫促進劑包括脂多醣體(lipopolysaccharide; LPS)與細胞素對吳郭魚頭腎巨噬細胞功能調控機制。結果顯示當添加γ-干擾素(interferon-γ)和腫瘤壞死因子-α (tumor necrosis factor-α;TNF-α)時,均會顯著地增進頭腎巨噬細胞對Streptococcus iniae的吞噬能力。同時當添加TNF-α、IFN-γ、白血球中介質(interleukin)-1β(IL-1β)或白血球中介質-2 (IL-2)作用15分鐘後,均可大幅提升頭腎巨噬細胞的超氧化物(superoxide)產量,然而於作用15分鐘後,細胞之superoxide產量則急速往下降。另外,抑菌能力實驗結果發現,當添加100 U/mL和1,000 U/mL IFN-γ後,確實可顯著地增加頭腎巨噬細胞的抑制細菌能力。然而,此時再加入α/β-干擾素(IFN-α/β)則會降低經100 U/mL IFN-γ與10 µg/mL LPS刺激過之頭腎巨噬細胞的抑菌能力。研究結果發現添加適當濃度之白血球中介質-4(IL-4)和白血球中介質-10(IL-10)則會降低經IFN-γ刺激後之頭腎巨噬細胞的抑菌能力。由此可知,TNF-α、IFN-γ、IL-1β及IL-2會增進細胞免疫力,而IFNα/β、IL-4及IL-10則會降低此能力。 |
英文摘要 | The aim of this research was to study the effects of lipopolysaccharide and various cytokines on the functions of tilapia head kidney-derived macrophages. The results revealed that the phagocytic ability of macrophages increased after stimulated with interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α). The superoxide production of macrophages was greatly enhanced with TNF-α, IEN-γ, interleukin-1β (IL-1β) or interleukin-2 (IL-2) added within incubation for 15 mins. After incubation for 15 mins, the superoxide production of macrophages was rapidly decreased. Furthermore, tilapia head kidney-derived macrophages treated with 100 U/mL and 1,000 U/mL IFN-γ significantly expressed a high bactericidal activity against Streptococcus iniae. In contrast, the bactericidal activity of the tilapia head kidney-derived macrophages stimulated with IFN-γ was inhibited by the addition of interleukin-4 (IL-4), interleukin-l0 (IL-10), and interferon-α/β. According to these results, TNF-α, IFN-γ, IL-1β, and IL-2 could enhance cellular immunities but IFN-α/β, IL-4, and IL-10 did not possess the abilities. |
