中文摘要 | 本研究嘗試以高效液相層析法(high-performance liquid chromatography; HPLC)搭配螢光檢測器檢測中華絨螯蟹肝胰腺與性腺中殘留之歐索林酸(oxolinic acid; OA)。先將OA標準品以50mMPBS(pH7.4)連續稀釋,再分別將各濃度藥物加入完全均質之1g肝胰腺與精巢組織,經超音波水浴加入n-hexane去脂後離心,將水層液通過C-18固相萃取管柱(SPE)後進行高效液相層析。結果顯示,利用逆相層析管柱(Cos-mosil5C18-MS),配合移動相acetonitrile:20mMPBS, pH3(74:26,v/v),流速1mL/min,以激發波長335nm與散射波長378nm之螢光檢測器可精確地檢測兩組織中添加之OA。結果顯示組織中回收率介於85%-146%,變異係數(CV)平均值均低於2.2%。最低檢測極限為0.5PPb(S/n≧3)。分析經OA餵食7天後之螃蟹樣本,發現本方法的確可自肝胰腺與精巢中檢測出OA之殘留。 |
英文摘要 | We analyzed the residue of oxolinic acid (OA) in the hepatopancreas and gonad of the Chinese mitten crab using high-performance liquid chromatography (HPLC) coupled with fluorescence detector. A standard of OA was prepared by diluting with 50mM phosphate buffered saline (PBS, pH 7.4). One-gram samples of hepatopancreas and testis samples were obtained and completely homogenized prior to addition of spiked OA. Samples were transferred into an ultrasonic water bath then defatted with n-hexane and centrifuged. The water layer was passed through C-18 solid phase extraction (SPE) column. Extracted OA was determined by HPLC with a fluorescence detector (excitation/emission=335nm/378nm) and a Cosmosil 5C18-MS reverse phase column. A mixture of acetonitrile and 20 mM PBS, pH 3.0 (26:74) was used as the mobile phase at a flow rate of 1.0mL/min. Our results indicated that the recoveries of OA from both tissues were about 85%-146%. Meanwhile, the mean of coefficient of variation value (CV) was less than 2.2%. The limit of detection was 0.5ng/mL (s/n≧3). After analyzing the samples from Chinese mitten crabs fed with OA for 7 days, residual OA was detected in both hepatopancreas and testis. |
