中文摘要 | 本文介紹蛋白質體技術應用於肺沖提液(BALF)的蛋白質分析。肺沖提液是肺泡上皮襯液(ELF)樣本中最常見的一種液體,可直接呈現肺部支氣管的肺泡蛋白質組成,因此在肺部疾病的診斷上是非常重要的樣本來源。肺沖提液分析可證實肺部因疾病所產生的細胞間質與蛋白質表現變化,在呼吸道疾病病患所取得的肺沖提液之蛋白質表現亦提供疾病分子醫學的診斷證明。蛋白質體分析技術提供了生物樣品中蛋白質樣本的成份分離、身分鑑定與特性描述,並且能在肺沖提液樣本提供一整體性的分析方法。本文回顧對肺沖提液的各種蛋白質體分析方法,包括樣品準備、二維膠體電泳法、質譜分析和生醫資訊庫等的分析方法。這些技術能在蛋白質表現的微小變化上發現與疾病相關之可能病理機轉。最後我們並將對肺沖提液蛋白質體分析與其臨床上肺部疾病的相關研究做一回顧性介紹。 |
英文摘要 | This review emphasizes modern technologies employed in the proteomics analysis of bronchoalveolar lavage fluids (BALF). BALF is the most common fluid for sampling the components of the epithelial lining fluid (ELF) and reflects the protein composition of the pulmonary airways. It is an important diagnostic source to investigate cellular and molecular changes in the course of lung disorders. The pattern of soluble proteins in BALF obtained from patients at different stages of respiratory disorders may provide deeper insights in the molecular mechanisms of the disease. Proteomic analysis involves the systematic separation, identification, and characterization of proteins presented in biological samples. It offers the comprehensive analytical tools to analyze BALF proteins at a systems level. This paper reviews the variety of analytical methods that have been used for proteomics analysis of BALF, including sample preparation, two-dimensional polyacrylamide gel electrophoresis, mass spectrometry, and bioinformatics. These technologies enable discoveries of tiny changes of protein expression associated with pathogenesis. In this review we introduce the present state of the investigations of BALF proteome and its powerful contribution both to a better knowledge of the lung structure at the molecular level and to the study of lung disorders at the clinical level. |
