中文摘要 | 免疫系統常誘導發炎反應來對應外來病原,並誘使防禦細胞前來,參與的免疫細胞包括單核球與其特化之巨噬細胞等,由防禦細胞的吞噬與消化作用,來清除外來病原與受損細胞。我們利用吞噬細胞活性為篩檢平臺來評估市面上聲稱具有增強免疫及抗發炎之中草藥,利用人類或小鼠巨噬細胞株進行體外培養之篩選模式以中草藥,包括人參、黨參、黃耆、山藥、甘草、靈芝、絞股藍、板藍根、當歸、玉竹、山茱萸、肉蓯蓉、沙菀子、仙茅、冬蟲夏草、千觔拔、金門一條根 、野牡丹、刺五加、麥門冬、沙棘、穿心蓮等中藥。本計畫中使用流式細胞儀或盤式螢冷光分析儀作為快速分析之工具,首先以巨噬細胞株對於吞噬螢光標示之大腸桿菌之能力與發炎細胞激素、鈣離子、NO表現產生調節的單味藥。結果顯示,會增加吞噬活力之中藥有菟絲子、刺五加、千觔拔、仙茅、黃耆、玉竹、板藍根與黨參等,劑量效應範圍為10 ng/ml, 100 ng/ml, 1 µg/ml與10 µg/ml;其中菟絲子、刺五加及板藍根有明顯增加鈣離子攝入細胞,並呈劑量依存性關係(100 ng/ml, 1 µg/ml, 10 µg/ml);菟絲子、刺五加、板藍根與千觔拔藥材均會影響NO的釋出,但相較於脂多醣(LPS)刺激並不顯著;會明顯降低TNF-α表現之藥材有千觔拔與板藍根,會明顯降低IL-1表現之藥材有板藍根,會明顯降低IL-6表現之藥材有板藍根,綜合以上結果顯示,板藍根可有效增加吞噬能力,並降低發炎激素之釋出,對於板藍根藥材進一步之藥效與有效成份分劃之分析,對於開發增強免疫並能抑制發炎反應之作用上有其潛力。 |
英文摘要 | Phagocyte is the first line in immune defense system. It can phagocytosis the foreign pathogen, and induced inflammatory mediator infiltration to kill the pathogen. In this study, we screen 22 folk herbal medicines to monitor the drug effect in enhancement the phagocytosis in culture macrophage. Inflammatory mediator including nitric oxide (NO), TNF-α, IL-1, IL-6 can be used the monitor the inflammation effect. As we know, activated phagcytosis can induced inflammatory cytokines and nitric oxide production in LPS stimulation as a positive control. The results indicated, there are 8 medicines can enhanced phagocytosis in 10 ng/ml to 10 µg/ml stimulation, including seed of Cuscuta austras R. BROWN, root of Acanthopanax senticosus (RUPR. ET. MAXIM.) HARMS, root of Flemingia strobilifera (L.) BR. EX AIT, rhizoma of Curculigo orchioides GAERTN, root of Astragalus membranaceus var. mongholicus (BGE.) HSIAO, rhizoma of polygonatum odoratum (MILL.) DRUCE, root of Isatis indigotica FORT and root of Codonopsis pilosula (FRANCH.) NANNF. But there NO release and inflammatory cytokines including TNF-α, IL-1, and IL-6 secession was minor increase and very significantly lower than LPS stimulation (P<0.001). Take all these data together, these folk herbal medicines can stimulate phagocytosis in different ranges of concentration, but they play a non-significant inflammatory mediators production, and might be a choice for enhancement the innate immunity medication. |
