| 中文摘要 | 在過去的這一年度,本計畫已完成50種中草藥或是臺灣特有植物成分對於人類腫瘤細胞的抗癌活性分析。研究結果發現數種成分具有抗癌活性,其中又以臺灣嘉賜樹(Casearia membranasea)所純化的10種化合物具有不錯的抗癌活性,它們依序為Casemembrin A-F、[rel-(2S, 5R, 6R, 8S, 10R, 18S, 19R)-diacetoxy-18-19-epoxy-6-hydroxy-2-(2-methylbutanoyloxy)cleroda-3, 13(16), 14-triene]及JCM-9、-10及-12,他們是一些新的clerodane雙萜類化合物。以TUNEL-reaction technique的實驗方法發現,它們皆可有效的促使腫瘤細胞進行凋亡作用,其半致死濃度(IC[9274])分別為1.2、24.2、0.5、1.4、1.9、2.6、2.1、1.8、13.7及>30µM,由結果也可看出Casemembrin C是其中最強的一個成分。因此,我們也積極的探討其抗癌作用機轉。我們從內在性及外在性的細胞凋亡路徑著手(Intrinsic and extrinsic apoptosis pathways),結果發現Caseamembrin C能夠有效的引發caspase-8、-9及其下游的casepase-3的活化,這些caspases的活化皆可促使腫瘤細胞進行凋亡作用,經深入研究發現內在性細胞凋亡路徑是主要的作用路徑。 |
| 英文摘要 | We have done a screening test on the examination of anticancer effect in various components from Chinese herbal medicines and plants in Taiwan. In this project, we have screened 50 pure compounds and found several active components. Among these compounds, a series of clerodane diterpenes (Caseamembrin A-F, [rel-(2S, 5R, 6R, 8S, 10R, 18S, 19R)-diacetoxy-18-19-epoxy-6-hydroxy-2-(2-methylbutanoyloxy)cleroda-3, 13(16), 14-triene, JCM -9, -10 and 1 12] from Casearia membranasea are effective anticancer agents. The anticancer IC[9274] values are 1.2, 24.2, 0.5, 1.4, 1.9, 2.6, 2.1, 1.8, 13.7 and 30 µM, respectively. The data showed that casemambrin C is the most effective one. Both the extrinsic and intrinsic apoptosis pathways are investigated in PC-3 cells responsive to caseamembrin C. The data demonstrated the involvement of activation of several casepases, such as caspase-8, -9, and -3. After a further analysis, it showed that the intrinsic apoptosis signaling was the predominant pathway. It is suggested that these clerodane diterpenes are potential anticancer agents for further development. |
