中文摘要 | 等鞭金藻 (Isochrysis galbana) 含有豐富的DHA,可作為輪蟲、橈足類及魚蝦貝類幼苗的餌料,惟其在高密度培養時,可能會面臨CO2供應不足的問題。本研究即在探討以小容量 (0.5 L 或 1.0 L) 靜置培養及大容量 (100 L) 打氣培養時,調整pH值或供應CO2對等鞭金藻增殖之影響。小容量靜置培養共分為四個試驗。在添加CO2或鹽酸 (HCl) 調整初始pH值之試驗中,以添加CO2調整pH 為6.5組者的生長最好,調整為5.5組者,無論是添加CO2或HCl,其增殖速率均比不添加之對照組為差。每日添加CO2或 HCl 將pH值調整為6.5之試驗中,以CO2組的增殖情形最佳,為對照組的221%;其次為HCl 組,而以對照組居末。添加碳酸氫鈉 (NaHCO3) 之試驗,以添加1 g/L組的細胞增殖情形最差,其餘五組(0.01 g/L、0.05 g/L、0.1 g/L、0.5 g/L與對照組)則差異不大。添加0.5 g/L NaHCO3,同時將pH值調整為6.5組,其增殖情形較僅添加NaHCO3者為佳,為對照組的212%。大容量打氣培養中,利用pH控制儀調控CO2或 HCl,使pH值維持於設定值,結果添加CO2調控pH值在7.5、7.0和6.5之三組間的細胞增殖情形無明顯差異,均較以HCl控制為7.5組和對照組為佳;而HCl控制組的效果則不比對照組好。CO2控制組在第7天細胞數最佳可達 1350 x 104 cells/ml,約為對照組的199%。本研究顯示,等鞭金藻在小容量靜置培養時,可添加濃度為0.5g/L的 NaHCO3,將pH調整為6.5;大容量打氣培養時,則可利用CO2控制藻水的pH為7.5,均可提高收獲時的藻細胞濃度。 |
英文摘要 | Being rich in Docosahexaenoic acid (DHA), the microalga Isochrysis aff. galbana (TISO) is regarded as a good live food for rotifer, copepod and oyster larvae, and as an indirect food for grouper larviculture. In order to resolve the insufficiency of CO2 with high-density algal culture, the effects of pH adjustment or CO2 addition for small volumes (0.5 or 1.0 L) of stagnant culture and large volume (100 L) of aerated culture under continuous illumination were studied. For the small-volume culture, CO2 or HCl was initially added to adjust the pH value to 5.5, 6.5, and 7.5. The growth was best for the CO2-6.5i group, while it was worst for the CO2-5.5i group. In the daily pH adjustment experiment, the CO2-6.5e group had the best performance with 221% proliferation compared to the control. The lowest cell density was observed in the culture with 1.0 g/L NaHCO3, and no significant difference was detected among the other groups. The addition of 0.5 g/L NaHCO3 and the adjustment of the pH to 6.5 with HCl resulted in 212% proliferation in contrast to the control. For the large-volume culture, CO2 gas or an HCl solution was used to maintain the pH at 6.5, 7.0, or 7.5. The addition of CO2 was more effective than HCl. However, there were no significant differences among the groups to which CO2 was added. The cell density reached 1.35 x 106 cells/ml, which was nearly twice that of the control on day 7. This study revealed that the addition of the carbon source can enhance the growth of I. galbana. |
