中文摘要 | 將甜瓜(金輝品種)之離葉於透明塑膠盒內以葉柄浸漬養液的方式培養,於28~32℃栽培6~10days後可由葉柄長出較多的根以維持葉片活力。以吹落孢子的方式將人工培養的白粉病菌Podosphaera xanthii分生孢子接種於已長根之離葉上,爾後培養於24℃/20℃(Day/night)光照12hrs之定溫箱,接種葉片發病面積隨接種濃度與接種時間之增加而增加。接種濃度30與100 spores/平方公分之處理4days後即可以肉眼觀察到病斑,並於12days後發病面積均達90%;若以接種濃度10spores/平方公分之處理經12days後發病面積約為60%。本病之發病適溫為20與24℃,12℃的環境下,接種18days後才能造成離葉發病面積達50%以上,而8℃以下與32℃以上接種21days仍未見病斑產生。同一植株不同位葉經盒栽離葉接種檢測,彼此發病面積呈不顯著差異,但下位葉較易黃化不適合作為盒栽離葉接種。以10spores/平方公分之接種源濃度於24℃/20℃(Day/night)光照12hr接種條件下,以盒栽離葉接種法測定甜瓜白粉病菌於甜瓜(金輝、秋蜜)、胡瓜(彩綠二號)、越瓜(銀華)、南瓜(仙姑)、蒲瓜(春鶯)等瓜類葉片的白粉病表現,發現各種瓜類均受感染,但以蒲瓜(春鶯)發病面積為42.5%,明顯較其他瓜類發病面積低(62.5~72.5%)。 |
英文摘要 | A cultured leaf method was developed for disease assessment of melon powdery mildew caused by Podosphaera xanthii in this study. The detached leaves of melon (Orient sweet- Golden Light) with petiole were cultured in the transparent plastic boxes which contained nutrient solution. After incubation for 6 to 10 days, the growth of roots and vigor of leaves were better at 28-32℃ in comparison with other temperatures. When leaves were inoculated with different concentrations of conidia of P. xanthii, and then incubated at 24℃/20℃ (day/night) with a 12 hr photoperiod, percentage of lesion area increased with increasing inoculum density and incubation time. The lesion area reached 90% after 12 days at the inoculum density of 30 or 100 spores/cm^2. However, the lesion area was about 60% at 10 spores/cm^2 after the same period of time. With this inoculation method, powdery mildews grew well at 20-24℃, but failed to grow at 8℃or 32℃. The disease development on cultured leaves of different ages was similar, but the lower leaves usually turned yellow during the inoculation period. The cultured leaf method was applicable to other cucurbit plants. With the inoculum density of 10 spores/cm^2 at 24/20℃ for 12 days, the lesion area of bottle gourd (Trendy) was 42.5%, lower than those (62.5-72.5%) of melon (Golden Light, Autumn Sweet), cucumber (Chailue No.2), pickling melon (Silver Charm) or squash (Fairy). |
