中文摘要 | 分子態之麴酸及維他命B₃和離子態之維他命C葡萄糖苷較易擴散至透析膜內而有利於透析程度的提高。以本研究所開發之線上微透析取樣技術結合高效液相層析系統,在0.020 M磷酸鹽緩衝溶液,甲醇體積百分率為40%,沖提液之pH5.5,Hypersil Fluophase PFP管柱之逆相層析4.5分鐘內即可完成上述三種美白劑的分離。測得濃度線性範圍是0.2~900 µM(維他命C葡萄糖苷)、0.5~2000 µM(麴酸)、0.2~4000 µM(維他命B₃),線性相關係數是0.9982、0.9999、0.9999,偵測極限是0.03、0.07、0.06 µM。基於真實樣品之分析物不是微量及偵測波長之強韌性的考量下,本實驗選擇254 nm做為後續分析之偵測波長。測得添加回收率介於92~106%,變異係數介於3.9~8.7%。本研究將所開發之線上微透析取樣技術結合高效液相分析系統,應用在六種真實樣品中之美白劑含量測定。確信本系統分析法之高準確度與可接受度。本系統是一簡單、可靠、快速之分析方法,它可同時測定不同劑型美白化妝品中維他命C葡萄糖苷、麴酸及維他命B₃。 |
英文摘要 | The results indicate that if Kojic acid (KA) and Niacinamide (VitB₃) are in the molecular form and Ascorbyl glucoside (AA-2G) is in the ionization species, they diffuse through the membrane more easily. AA-2G, KA, and VitB₃ were well separated less than 4.5 min on a reversed-phase Hypersil Fluophase PFP column eluted with 0.020 M phosphate-buffer solution in 40% (v/v) methanol at pH 5.5. Calibration curves were found to be linear in the 0.2-900 μM (AA-2G), 0.5-2000 μM (VitB₃). The correlation coefficients of linear regression analysis were with the range 0.9982-0.9999. The detection limits of these compounds are 0.03 μM for AA-2G, 0.07 μM for KA, and 0.06 μM for VitB₃. Considering the robustness of detection wavelength and the levels of the analytes in the samples are high (0.1%-2%). The analytes were all detected by ultraviolet light absorption at the wavelength of 254 nm. The recovery values were in the range 92-106% with good reproducibility (R.S.D. = 3.98-8.7%). Six commercial available bleaching cosmetics were assayed using the procedure developed in this study. The results obtained confirm the precision of the method and confirm the label claim on the cosmetics. The proposed method provided a very simple procedure to determine AA-2G, KA, and VitB₃ in various dosage forms of bleaching cosmetics. |
