中文摘要 | 前列腺癌是我國最常見的男性生殖泌尿癌,也是發生率上升快速的癌症,大部分病因不明,其中一些與外來病毒(猿猴病毒40, simian virus 40 )嵌入宿主核酸有關。雙螺旋核酸中的空間稱為溝(groove),此處可與特定分子接合,以調控細胞製造所需物質,若此位置被其他物質占據或破壞,則癌細胞將被抑制生長或殺死。 本研究即在探討利用一特殊設計的單股核酸,與SV 40( simian virus 40 )核酸互補,此核酸一端與FAM 藥物接合,當它嵌入癌細胞雙螺旋核酸的大溝中,形成三螺旋核酸時,會近距離與雙螺旋核酸接觸,此時經藥物作用,會與癌細胞之雙螺旋核酸接合;而正常細胞與特殊設計的單股核酸無法形成三螺旋,與藥物無法近距離接觸,故不受藥物影響。本實驗結果發現三螺旋結構在沒有鎂離子下熔點為37℃,鎂離子濃度為10 mM時熔點為 55 ℃。在細胞外結合效率實驗結果為70 %。 此三螺旋核酸的結構可望應用在癌症基因治療,若其結合效率能提升,將有助於癌症病患治療,或者配合其他療法如放療或化療,應可提高癌症病患治癒率。 |
英文摘要 | Males in our country commonly suffer from prostate cancer. The occurrence rate of prostate cancer also increases rapidly. Most causes of this cancer are not clear although some of them relate to nucleic acid inset by foreign viruses. The space between double helix, groove, can be attached with specific molecules to make cell manufacturing demanded materials. In case the groove is occupied or damaged with other molecules, the cancer cells will be suppressed or killed. In this research, a designed single strain helix complementary to a carcinogenic virus SV40 was attached with FAM on one end. When the single strain inset the major groove of the cancer cell’s double helix to form a triplex strain helix, it could closely contact the helix. FAM was able to bind the double helix of the cancer cell. On the other hand, the normal cell was unable to form triplex strain with the designed single strain. Since FAM was not capable of closely contacting the double helix, it would not affect the normal cell. The results illustrated that the melting point of the triplex strain was 37℃ without Mg2+ ion, and 55℃ at 10mM of Mg2+ ions. The integration ratio outside the cell was 70﹪. The triplex strain helix was expectedly applicable in cancer gene therapy from this study. It will be useful in curing cancer patients if the integration ratio can be enhanced. By incorporating other therapies (e.g. radiotherapy or chemotherapy), the cure rate can be increased. |
