中文摘要 | 本研究進行微生物固定化相關研究,測試利用不同固定化載體進行ABE發酵生產丁醇的效率,結果發現固定化於磚頭載體的丁醇生產菌Clostridium acetobutylicum CPCNE菌體呈現長桿狀型態且活動力較佳,細胞活性維持在較高狀態,有利菌體生長及進行發酵作用。依葡萄糖利用速率結果推測磚頭、發泡沸石、海綿、玉米稈及棉為較佳的固定化載體選擇;依丁醇產量結果發現磚頭、海綿、不織布、發泡沸石、玉米稈及棉為較佳的載體選擇,其中以粒徑2~3.36 mm的磚頭粒子固定化效果最好,在48小時內即可產生近13 g/L丁醇,且丙醇與乙醇產量也較高。相較於其他載體,磚頭粒子(粒徑_2~3.36 mm)的微生物固定化發酵效果最佳,且磚頭粒子重量較重,在發酵反應器中易沉降,便於與液態溶劑產物分離,所以最適合作為固定化載體使用。 |
英文摘要 | In this study, the efficiency of butanol production by ABE fermentation in immobilized cell reactors using different carriers was discussed. The results showed that Clostridium acetobutylicum CPCNE immobilized in bricks performed long rod-shaped, high cell activity and better fermentation efficiency. By the assay of glucose-consumed efficiency, it was suggested that the brick, boiling chip, corn stalk and cotton were appropriate carriers for the immobilization of Clostridium acetobutylicum CPCNE. Similarly, by the assay of butanol-producing efficiency, it was suggested that the brick, nonwoven, boiling chip, corn stalk and cotton were better carriers for the immobilization of Clostridium acetobutylicum CPCNE. The cells immobilized in brick pieces (2~3.36 mm size) performed the best butanol-producing ability while the butanol produced by this immobilized cell reactor was about 13 g/L following 48 h of fermentation. Compared to other carriers, brick pieces (2~3.36 mm size) were the most suitable for immobilization of Clostridium acetobutylicum CPCNE because they could be easy to sink under the bottom of the bioreactor to separate from solvents produced by ABE fermentation. |
