中文摘要 | 本研究的主要目的是要瞭解Leptin蛋白質結構同時探討Leptin對於食慾調節和體重控制的機轉。Leptin是一種16-K Da的賀爾蒙,是屬於Cyotokine家族的一員;早期動物實驗指出Leptin可以消除肥胖,另外更有研究指出Leptin可以對大腦產生減少食慾的信號;它一方面可以阻止神經胜_Y的釋放(促進食慾的神經介質),另一方面可以促進α黑細胞激素賀爾蒙的釋放(減少食慾的神經傳遞質)。但近期的臨床實驗指出。如果注射Leptin到肥胖者的體內,會導致過敏;而且大多數肥胖者都帶有正常的Leptin基因和正常的血液Leptin濃度,當成肥胖的原因仍有待進一步的研究,減肥並不是只要增加體內的Leptin濃度即可,因此Leptin並不是調節食慾或體重控制的仙丹妙藥。 |
英文摘要 | Leptin, derived from the Greek root lept_s (meaning thin), is a 16-lD hormone□ involved in the regulation of appetite and body weight. This hormone is secreted mainly from white adipose cells or fat-storage cells, and the amount of its secretion increases as the body fat accumulates.20 The earlier discoveries of the weight-reducing effects of leptin□have created tremendous sceientific interest in unraveling the mechanisms of action of leptin in regulating body weight. This has given new hope for solving the human obesity problem. A large body of research has focused on how leptin acts as a satiety factor to regulate appetite and body weight within the brain.□ Less research, in comparison, is devoted to investigating the weight-reducing effect of leptin through its interference with lipid metabolism in the body.□ This review review aims to introduce the structure of leptin, some earlier experiments suggesting the role of leptin as an appetite and body weight regulator, some proposed mechanisms of action of leptin, and the success and failures of leptin therapy. |
