中文摘要 | 本研究首先探討培養液中不同氮源成份及有無葡萄糖對Lactobacillus acidophilus生長的影響,並評估初菌量及培養時間對L. acidophilus於培養液中生長之情形。以牛肉萃(beef extract)及casitone為氮源之L. acidophilus在37℃培養48小時之菌數均比酵母萃(yeast extract)培養液為佳,當有葡萄糖存在時,L. acidophilus的生長情形較優。於含葡萄糖之牛肉萃培養液的L. acidophilus在10[feb4]CFU/mL初菌量經培養24小時後的菌數顯著高於對照組;初菌量為10[fed5] CFU/mL時,於含葡萄糖之牛肉萃培養液中培養18小時後,L. acidophilus生長亦較優。至於在10[8fb6] CFU/mL初菌量,於有或無葡萄糖培養液的菌數變化無顯著差異(p>0.05)。另外,L. acidophilus於有或無葡萄糖之casitone培養液在36小時培養期間,菌數亦無明顯差異(p>0.05)。由,L. acidophilus在培養液的生長情形發現,理想的生長條件包括接菌量10[fed5] CFU/mL、牛肉萃為氮源、葡萄糖為碳源及培養時間為24小時,並依此作為後續篩檢抗性澱粉及其影響益生菌生長的基礎培養條件。 |
英文摘要 | The purpose of this study was to investigate the effects of additional different nitrogen sources and glucose on the growth of Lactobacillus acidophilus. Factors, including initial cell count and incubation time on the growth of L. acidophilus in the tested broths were evaluated. Results showed that maximum cell counts of L. acidophilus were obtained when the bacteria used beef extract or casitone as a nitrogen source. The same result was also observed in the broth with glucose added. L. acidophilus grown in glucose-containing beef extract broth for 24 h. with an initial cell count of 10(feb4) CFU/mL, resulted a significantly higher cell count compared to the control. The same result was also obtained when L. acidophilus was incubated in glucose-containing beef extract broth after 18h. The cell counts of L. acidophilus incubated in broths with or without glucose had no significant difference (p>0.05) as initial cell count of 10(8fb6) CFU/mL was used. In addition, casitone broths with or without glucose added did not affect the cell counts of L. acidophilus after a 36 h-incubation. The present data indicated that the optimal growth conditions for probioties were as such, 10(fed5) CFU/mL initial cell count, beef extract as the nitrogen source, glucose as the carbon source and 24 h-incubation, respectively. |
