中文摘要 | 本研究目的在探討臺灣原生藥用植物-高氏柴胡(Bupleurum kaoi Liu, Chao et Chuang)經由組織培養大量繁殖種苗的可行性。試驗結果顯示,將腋芽培養於含有1 mg/l Benzyladenine (BA)配合0.1 mg/l α-Naphthaleneacetic acid (NAA)之1/2MS基本鹽類培養基,可建立高氏柴胡之無菌培養。將此無菌苗培養於添加0.25 mg/l BA之1/2MS基本鹽類培養基可達到大量繁殖的目的,然而部份芽體卻形成玻璃質化現象;組織培養苗培養於含有0-0.8 mg/l kinetin之1/2MS培養基則可避免玻璃質化苗產生,但繁殖倍率較BA(0.25-2 mg/l)處理低。基本鹽類濃度以1/2MS培養基對於高氏柴胡芽體增值較佳;蔗糖則以3%最適合。液態培養下正常苗繁殖率低於固態培養,且培養6週後產生嚴重的玻璃質化問題;以液態培養基預培養2-14天後再移至固態培養基,繁殖倍率仍無法高於固態培養,因此液態培養方式並不利於高氏柴胡組培苗之增殖。 |
英文摘要 | An efficient method for in vitro shoot multiplication of Bupleurum kaoi Liu, Chao et Chuang using the axillary bud explants has been developed. The axillary buds dissected from nodal segments of the field grown plants induced shoots after culturing for 4 weeks on half-strength MS medium supplemented with 1 mg/l benzyladenine (BA) in combination with 0.1 mg/l α-naphthaleneacetic acid (NAA). Among the various concentrations of BA (0-2 mg/l) or kinetin (0-0.8 mg/l) tested, maximum shoot multiplication (7.3 shoots per explant) was achieved in the in vitro grown shoots by culturing on half-strength MS medium supplemented with 0.25 mg/l BA for 8 weeks. Although BA was more effective than kinetin in shoot multiplication, BA stimulated hyperhydric shoots (10-33%) after 6 weeks of culture. Prolonged culture period from 6 weeks to 8 weeks resulted in higher number of hyperhydric shoots (16-47%). Half-strength of MS basal medium and 3% sucrose as carbohydrate source was found optimum for shoot multiplication. Pretreatment of the explants by culturing them on liquid half-strength MS basal medium containing 0.25 mg/l BA for 2-14 days did not support shoot multiplication. |
