中文摘要 | 炭疽病菌(Colletotrichum spp.)是台灣地區普遍存在的真菌之一,也是農業上重要的一屬植物病原真菌;由於它的種類繁多,正確而快速的分類鑑定,對於此真菌的研究、利用或防治相當重要。本研究利用台灣地區23株炭疽病菌菌株,以PCR CITS1及CITS4引子對,增幅各菌株之核糖體核酸(ribosomal DNA, rDNA)之內轉錄間隔區(internal transcribed spacer, ITS)(ITS1-5.8S-ITS2),再以7種限制?Bam HI、Bst UI、Eco RI、Msp I、Pvu II、Sau 3AI和Scr FI剪切,可得到各菌株呈現不同型式之核酸片段。以Bst UI可產生最多種的核酸條帶型式,共有12種;其次為Scr FI和Msp I兩種限制?,分別產生10種類型的條帶組合;Eco RI及Sau 3AI有9種;Bam HI有8種;Pvu II所產生的條帶類型最少,只有7種。以Bst UI、Pvu II 和Eco RI三種限制?所產生的多型性互相搭配所得到的區分效果最佳,依此限制片段長度多型性(restriction fragment length polymorphism,RFLP)圖譜,可將23株供試炭疽病菌菌株,區分成20種核酸條帶類型。另一方面藉由樹狀關係圖,探討各菌株間之親疏關係。 |
英文摘要 | The internal transcribed spacer (ITS) region of ribosomal DNA (rDNA) has proven useful as a molecular marker to identify fungal species, including species of Colletotrichum . Twenty-three Colletotrichum isolates, which were isolated in Taiwan and used in this study were obtained from the Bioresources Collection and Research Center (BCRC). PCR products were generated from the region spanning the ITS and the 5.8S rRNA gene with primer pairs, CITS1 and CITS4, which correspond to the conserved sequences of the rDNA. Seven restriction endonucleases were used to digest the PCR products in order to generated restriction fragment length polymorphism (RFLP). The RFLP pattern derived from Bst UI, Pvu II, and Eco RI led to identification of the greatest number of species; that is, 20 classified types were recognized among 23 Colletotrichum isolates. Clustering analysis of these 23 isolates was also conducted based on the RFLP produced by those 3 restriction enzymes. |
