中文摘要 | 本研究旨在探討大雪山台灣肖楠種子園開花旺盛營養系(no. 11)與開花稀疏營養系(no. 22)經激勃素A3 (GA3)處理後之開花效果,並定量分析在不同時期採收之芽體內各種不同激勃素的含量變化。以未經GA3處理之上述二個營養系和六龜種子園11號營養系作為對照組。經GA3處理之no. 11營養系開花比率99.7%,no. 22營養系85.6%,對照組六龜11號營養系7.1%,另二個未經處理者沒有花芽分化。激勃素定量分析在1999年5月下旬GA3處理之後,以間隔1及3 wk切取芽體頂端1-2 mm冷凍乾燥,秤取0.5 g乾重萃取分析激勃素含量的變化。結果GA3處理之樣品測到大量GA1與GA3及微量的GA20,1 wk後no. 11營養系GA1為920 ng˙g-1dry weight(dw)與GA3為1,200 ng˙g-1dw,no. 22營養系GA1為1,100 ng˙g-1dw與GA3為1,312 ng˙g-1dw。三個星期後no. 11營養系GA1為436 ng˙g-1dw與GA3為600 ng˙g-1dw,no. 22營養系GA1為1,118 ng˙g-1dw與GA3為826 ng˙g-1dw。兩個時間點的GA20含量過低無法定量。由GA1和GA3含量的變化,顯示no. 11營養系1和3 wk間濃度下降較大,而no. 22營養系濃度變化較小。由於芽體發現有GA24存在,證實台灣肖楠激勃素的合成路徑走non-13-hydroxylation pathway途徑,而激勃素的代謝速率影響花芽的分化。 |
英文摘要 | The flowering responses of GA3-treated branches of good flowering clone no. 11 and poor flowering clone no. 22 of Calocedrus formosana grown at Tashueshan were investigated. Quantitative analysis of gibberellins in buds harvested at different developmental stages was conducted as well. Samples collected from GA3-untreated clones grown at Tashueshan and clone no. 11 grown at Liukuei were used as controls. Flowering percentages of clone nos. 11 and 22 treated with GA3 were 99.7% and 85.6%, respectively. No flowers were found in the control clones of nos. 11 and 22, except for clone no. 11 grown at Liukuei which had a 7.1% flowering rate. Buds were collected at the 1st and 3rd weeks after branches were treated with GA3 in late May 1999. The tips of buds 1-2 mm long were excised and freeze-dried. Gibberellins extracted from 0.5 g of dry bud tips were quantified. Large amounts of GA1 and GA3 and minimal GA20 were detected in GA3-treated samples. The amount of GA1 and GA3 derived from clone no. 11 samples collected at the 1st and 3rd weeks were 920 and 1,200 ng˙g-1dry weight(dw), and 436 and 600 ng˙g-1dw, respectively, while those from clone no. 22 were 1,100 and 1,312 ng˙g-1dw, and 1,118 and 826 ng˙g-1dw, respectively. The amounts of the decrease in GA1 and GA3 from the 1st to the 3rd week were greater in clone no. 11 than in clone no. 22. We failed to detect a minimal amount of GA20. Minimal GA24 was detected, suggesting that the biosynthesis of gibberellins might proceed through a non-13-hydroxylation pathway. |
