中文摘要 | 為瞭解不同種類、濃度之生長素對不同部位枝尖培養體胚形成及植株再生的影響,以栽種甘藷臺農67號(TNG67)及臺農68號(TNG68)之不同節位之預芽及側芽之枝尖為培植體,接種於MS及經修飾之MS(代號MMS),添加60 g/l sucrose,10.8 g/l agar之基礎培養基,再分別添加不同度(0.1,0.5, 1 mg/l)之2,4-D或4-FA之12 種培養基,暗處理6~8週,進行胚性癒合組織之誘導,之後再繼代於原來之培養基,或含有0.5 mg/l ABA之MSA培養基中進行體胚分化及芽體再生之誘導,再將再生之芽體繼代於含有0.1mg/l IAA之MSI培養基中誘導芽體發根,結果如下: 甘諸臺農67號頂芽與側芽枝尖之月性癒合組織誘導率低於臺農68號。在含有0.1,0.5 mg/l auxin之培養基中,二品種中上節位之胚性癒合組織誘導率顯著優於下節位;對於胚性癒合組織誘導,培養基中添加4-FA之效果優於2,4-D,其中以0.5mg/l 4-FA之誘道效果最佳;MMS基礎培養基對胚性癒合組織誘導未必優於以MS為基礎培養者;將胚性癒合組織繼代培養於含有0.5 mg/l ABA之培養基中體胚分化形成芽體,再繼代培養於含有0.1mg/l IAA之培養基可誘導芽體發根長成小植株,健化後移植可正常生長。 |
英文摘要 | Sweet potato, cultivars TNG67 and TNG68, were used to study somatic embryogenesis. Various combination of basal media and plant hormones were tested in the experiments. Under darkness, embryogenesis callus were induced from the apical and lateral shot tips of in-vitro-cultured plantlets. After 6 to 8 weeks, the callus were transferred to MS or MMS (modified MS) media containing 0.5 mg/l ABA for somatic embryo indnction. Comparing the cultivars, TNG68 induce more embryogenic callus than TNG67. Comparing the tissues cultured under low auxin condition (0.1-0.5 mg/l), the apical explants and lateral explants closed to the apical explants, generate more embryonic callus than those away from the apical explants. Comparing the effect of auxins, media containing 4-FA induce more embryonic callus than 2,4-D, where 0.5 mg/l 4-FA appeared to be the optimal concentration. There were no obrions difference between MS and MMS basal media. The transferring of embryonic callus from induction media to media containg 0.5 mg/l ABA, appeared to improve the induction of somatic embryo. |
