英文摘要 | The present review focuses on investigations which implicate a unique role of κ-opioid receptors in dependence upon and withdrawal from butorphanol. Evidence will be summarized to indicate that brain κ-opioid receptors are more directly involved than are µ-opioid receptors ni the development of physical depedence upon, and the expression of withdrawal from, butorphanol, as opposed to the prototypical opioid analegesic, morphine. Quantitative analyses of κ-opioid receptors suggest than internalization of receptors and/or de novo synthesis of receptors occur differentially between rats treated with butorphanol and those treated with morphine. Regionally-specific expression of brain κ-opioid receptor subtypes during the development of butorphanol dependence and withdrawal in rats has also been demonstrated. Finally, evidence indicates than butorphanol treatment causes rat brain κ-opioid receptors to develop supersensitivity to opioid receptor antagonists, possibly as a result of conformational change, that may explain enhanced responses to antagonist-precipitated withdrawal. Within the brain, the locus coeruleus has been identified as a primary site wherein glutamate, acting through the N-methyl-D-aspartate glutamate receptor, mediates the expression of behavioral responses during withdrawal from dependence upon butorphanol. The specific nature, location and synaptic function of κ-opioid receptors within the locus coeruleus play important roles in regulation of glutamatergic neurotransmission and contribute facets to the development of dependence upon and withdrawal from butorphanol. |
