英文摘要 | Opioid agonists have been used widely and successfully in pain management paradigms for centuries. Though they are among the best medicines for pain control, their many side effects have diminished the effectiveness of these drugs. One of these side effects is the development of tolerance and dependence during chronic administration. In order to develop better pain treatment paradigms and an ideal opioid analgesic agent, the molecular mechanism for the development of these side effects should be elucidated. Hence, in this review, we examine the various hypotheses on the molecular mechanisms of opioid tolerance development. We will summarize the experimental data supporting the role receptor desensitization as the mechanism for in vivo opioid tolerance development. We will discuss the roles of agonist-induced receptor phosphorylation, receptor internalization and the role of arrestin interaction on the opioid receptor desensitization. The significance of receptor desensitization in the overall unifying mechanism of opioid tolerance development will be discussed. |
